目的:观察复方烫疡油(Fu fang tang yang you,FTY)促进小鼠创面愈合、减少瘢痕形成的作用,并探讨其作用机制。方法:采用小鼠深Ⅱ0烫伤模型。创面外用复方烫疡油治疗,并用表皮生长因子(Recombinant human epidermal growth factor,rhEGF)、磺胺嘧啶银霜、芝麻油(赋形剂)作疗效对比,用生理盐水作空白对照。测定创面组织创伤后8、12、24、48、72h干湿重比例以了解组织创伤后水肿情况;透明膜描绘-求积议测定创面伤后3、7、14h创面愈合情况;并用免疫组织化学法观察Ⅰ、Ⅲ型胶原、TGF-β1(Transforming growth factor-β1,TGF-β1)的变化;TUNEL法定量观察组织细胞的凋亡指数;原位杂交观察胶原酶(Marix Metalloproteianse1,MMP-1)及金属蛋白酶抑制剂(Tissue inhibitor of metalloproteinase-1,TIMP-1)mRNA的表达。结果:复方烫疡油能减轻创面炎症,降低创伤组织水肿,改善Ⅰ、Ⅲ型胶原比例,减少瘢痕形成,促进创面愈合。结论:复方烫疡油具有抗炎、抗感染,促进创面愈合,减轻瘢痕形成的作用。
OBJECTIVE: To observe the effect of Fufangfangyangyou (FTY) on wound healing and scar formation in mice, and to explore its mechanism of action. Methods: Mouse deep II0 scald model was used. The topical wound was treated with compound iron-burning oil, and epidermal growth factor (rhEGF), sulfadiazine silver cream, and sesame oil (excipient) were used for comparison of efficacy. Normal saline was used as a blank control. The proportion of dry and wet weights at 8, 12, 24, 48, and 72 hours after wounding was measured to understand the edema after tissue trauma; the wound healing at 3, 7, and 14 hours after wound healing was measured by transparent film delineation-integration; and immunohistochemistry was performed. Methods The changes of type I and type III collagens and transforming growth factor-β1 (TGF-β1) were observed. The apoptotic index of tissue cells was quantified by TUNEL method. The collagenase (Marix Metalloproteianse1, MMP-1) was observed by in situ hybridization. And metalloproteinase inhibitor (Tissue inhibitor of metalloproteinase-1, TIMP-1) mRNA expression. Results: Compound scald oil can reduce wound inflammation, reduce wound tissue edema, improve the proportion of type I and III collagen, reduce scar formation, and promote wound healing. Conclusion: Compound scald oil has anti-inflammatory, anti-infection, promote wound healing and reduce scar formation.
