目的探究特异性蛋白3(Sp3)与三磷酸腺苷结合蛋白G亚族成员2(ABCG2)在人结肠癌组织中的表达及其相关性,并探讨其临床意义。方法收集2010年1月1日至2015年1月1日普通外科行外科手术治疗的50例患者的结肠癌组织及对应癌旁组织,采用免疫组化染色检测Sp3和ABCG2蛋白在组织中的表达,采用Spearman相关性分析两种蛋白在结肠癌组织中表达的相关性,分析肿瘤组织中Sp3和ABCG2蛋白表达的临床意义。结果Sp3和ABCG2蛋白均高表达于结肠癌组织中,其阳性表达率分别为50.0%和66.0%,明显高于癌旁组织中Sp3(20.0%)和ABCG2蛋白(26.0%)阳性表达率(P均<0.01)。临床资料分析发现,有淋巴结转移及TNM分期(Ⅲ+Ⅳ期)的结肠癌组织中Sp3及ABCG2蛋白阳性表达率明显高于无淋巴结转移和Ⅰ+Ⅱ期的结肠癌组织(P<0.01,P<0.05);相关性分析表明,Sp3和ABCG2蛋白在结肠癌组织中的表达呈显著正相关关系(r=0.801,P<0.01)。结论结肠癌组织中Sp3和ABCG2蛋白表达上调,且与患者恶性临床特征相关,二者表达的正相关关系提示在结肠癌中ABCG2可能是Sp3的转录靶点之一。
Objective To investigate the expression and correlation of specific protein 3 (Sp3) and adenosine triphosphate binding G subfamily member 2 (ABCG2) in human colon cancer and its clinical significance. Methods Colorectal cancer tissues and corresponding paracancerous tissues from 50 patients who underwent surgery in the general surgery department from January 1, 2010 to January 1, 2015 were collected. The expression of Sp3 and ABCG2 in tissues was detected by immunohistochemistry , Spearman correlation analysis of the two proteins in colon cancer tissue expression correlation analysis of tumor tissue Sp3 and ABCG2 protein expression in clinical significance. Results The positive expression rates of Sp3 and ABCG2 in colon cancer tissues were 50.0% and 66.0%, respectively, which were significantly higher than those in adjacent tissues (20.0%) and ABCG2 (26.0%) (P All <0.01). Clinical data analysis showed that the positive expression rates of Sp3 and ABCG2 protein in colon cancer tissues with lymph node metastasis and TNM stage (stage Ⅲ + Ⅳ) were significantly higher than those without lymph node metastasis and stage Ⅰ + Ⅱ (P <0.01, P <0.05). Correlation analysis showed that there was a significant positive correlation between Sp3 and ABCG2 protein expression in colon cancer tissues (r = 0.801, P <0.01). Conclusion The expressions of Sp3 and ABCG2 in colon cancer tissues were up-regulated, and correlated with the malignant clinical features of the patients. The positive correlation between the expression of ABCG2 and ABCG2 may be one of the transcriptional targets of Sp3 in colon cancer.
