目的建立测定丹参素在大鼠血浆中浓度的LC-MS/MS方法,并将该方法用于冠心舒胶囊在大鼠体内的药动学。方法色谱柱为C18柱(50 mm×2.1 mm,1.7μm),采用梯度洗脱的方式分离被测物质,利用液液萃取的方法处理血浆样品,以多反应监测(multiple reaction monitoring,MRM)扫描方式检测。结果丹参素在线性范围内线性关系良好,日内日间精密度(RSD)均小于15%,准确度(RE)在±15%以内,平均提取回收率在80%以上,基质效应在90%~110%。结论该方法适合于冠心舒胶囊在大鼠体内的药动学研究。
OBJECTIVE To establish a LC-MS / MS method for the determination of Danshensu in rat plasma. The method was applied to the pharmacokinetics of Guanxin Shu Capsule in rats. Methods The chromatographic column was C18 column (50 mm × 2.1 mm, 1.7 μm). The analytes were separated by gradient elution. The plasma samples were extracted by liquid-liquid extraction and analyzed by multiple reaction monitoring (MRM) Way to detect. Results Danshensu showed a good linearity within the linear range. The RSD was less than 15%, the accuracy (RE) was within ± 15%, the average extraction recovery was above 80%, and the matrix effect was within 90% 110%. Conclusion This method is suitable for pharmacokinetic study of Guanxin Shu capsule in rats.
