目的探究杞红方对寒凝血瘀衰老大鼠心脏的影响。方法 48只SD大鼠随机分为空白对照组,模型组,维生素E组(13.5 mg/kg),杞红方低、中、高剂量(45、90、180 mg/kg)组。除空白对照组外,其余5组运用28 d臭氧致衰复合14 d冰水致寒凝血瘀造模法,制作寒凝血瘀衰老大鼠模型。第29天腹主动脉取血分离血清、血浆,分离心脏,检测血清总超氧化物歧化酶(T-SOD)、铜锌超氧化物歧化酶(Cu-Zn-SOD)、锰超氧化物歧化酶(Mn-SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)的活力,丙二醛(MDA)含量,血浆凝血4项(PT、APTT、TT、FIB)。通过HE染色观察心脏细胞形态,Masson染色观察纤维沉积量,免疫组化检测转化生长因子β1(TGF-β1)表达量以评价寒凝血瘀衰老模型,并观察杞红方对大鼠心脏的保护作用。结果与空白对照组相比,模型组T-SOD、Cu-Zn-SOD、Mn-SOD、CAT、GSH-Px活力明显降低(P<0.05),MDA含量明显升高(P<0.05),PT、APTT明显降低(P<0.05),FIB含量明显升高(P<0.05),光镜下Masson染色,形态计量心脏胶原纤维沉积明显增加(P<0.05),TGF-β1表达量明显升高(P<0.05)。与模型组相比,杞红方中、高剂量均能明显回升T-SOD、Cu-Zn-SOD、CAT、GSH-Px活力(P<0.05),明显回降MDA含量(P<0.05),明显回升PT、ATPP(P<0.05),明显回降FIB含量(P<0.05),明显回降心脏细胞胶原纤维沉积(P<0.05),明显回降TGF-β1表达量(P<0.05)。结论杞红方对寒凝血瘀衰老大鼠的心脏具有保护作用。
Objective To investigate the effect of QIHONGFANG on the heart of old rats with cold coagulation and blood stasis. The morphology of cardiac cells was observed by HE staining, the amount of fiber deposition was observed by Masson staining, the expression of transforming growth factor-β1 (TGF-β1) was detected by immunohistochemistry to evaluate the effect of QHP on rat heart . Results Compared with the blank control group, the activities of T-SOD, Cu-Zn-SOD, Mn-SOD, CAT and GSH-Px in model group were significantly decreased (P < (P <0.05). The amount of FIB was significantly increased (P <0.05). Masson staining under light microscope showed that the deposition of cardiac collagen fibers was significantly increased (P <0.05) and the expression of TGF-β1 was significantly increased P <0.05). Compared with the model group, Qihong recipe could significantly up-regulate the activities of T-SOD, Cu-Zn-SOD, CAT, GSH-Px and the MDA content of the model group (P <0.05) (P <0.05), significantly decreased FIB content (P <0.05), significantly decreased the deposition of collagen fibers in cardiac cells (P <0.05) and significantly decreased the expression of TGF-β1 (P <0.05). Conclusion Qi-hong Fang has the protective effect on the heart of old rats with cold coagulation and blood stasis.
