目的探讨血浆标本的保存温度与保存时间对胰岛素浓度的影响程度。方法取18位患者的血液,分离血浆并分成25份,保存于由不同温度和时间组合的25种环境条件下,分析其血浆胰岛素浓度与血浆标本的保存温度、保存时间的关系。结果①血浆的保存温度(F=4.0,P=0.023)与保存时间(F=27.3,P=0.000)对胰岛素浓度的影响均具有统计学意义,且二者对胰岛素浓度的交互影响具有统计学意义(F=7.3,P=0.001)。②4℃时,不同时间的胰岛素浓度差异无统计学意义(P>0.05),20℃和37℃时,不同时间的胰岛素浓度差异均具有统计学意义(P<0.05)。③4℃时,不同保存时间的血浆相对于即时冷冻血浆胰岛素浓度的偏离度呈小范围水平波动趋势;20℃和37℃时,随着时间增加,各标本胰岛素浓度的偏离度大都呈迅速下降趋势。结论血浆标本的保存温度与保存时间均是影响血浆胰岛素浓度的因素,低温是保持血浆胰岛素稳定性的最重要措施。
Objective To investigate the influence of preservation temperature and preservation time of plasma samples on insulin concentration. Methods The blood samples of 18 patients were collected and divided into 25 parts and stored in 25 kinds of environmental conditions with different temperature and time. The relationship between the plasma insulin concentration and the storage temperature and storage time of plasma samples were analyzed. Results ① The effects of plasma preservation temperature (F = 4.0, P = 0.023) and storage time (F = 27.3, P = 0.000) on the insulin concentration were statistically significant, and the interaction between the two on insulin concentration was statistically significant Significance (F = 7.3, P = 0.001). ② At 4 ℃, there was no significant difference in insulin concentration at different time points (P> 0.05). At 20 ℃ and 37 ℃, the difference of insulin concentrations at different time was statistically significant (P <0.05). (3) At 4 ℃, the deviations of insulin concentration in plasma at different storage times showed a trend of small-scale fluctuation. At 20 ℃ and 37 ℃, the deviations of insulin concentrations in most samples showed a tendency to decrease rapidly with the increase of time . Conclusion The plasma temperature and storage time are the factors affecting the plasma insulin concentration. Low temperature is the most important measure to maintain the stability of plasma insulin.
