目的 探讨热休克蛋白 70 (HSP70 )在局灶脑缺血再灌流后的变化和作用。方法 采用线栓法制备大脑中动脉缺血再灌流模型。应用免疫组化方法观察 HSP70的组织学分布 ,利用 RT- PCR方法检测缺血皮层与基底节区脑组织 HSP70 m RNA相对含量。结果 基底节区 HSP70阳性细胞较少 ,持续时间短 ,皮层区 HSP70阳性细胞较多 ,持续时间长。RT- PCR结果表明 HSP70 m RNA相对含量与免疫组化结果相一致。结论 HSP70是缺血神经元损伤的敏感指标 ,缺血皮层区 HSP70表达的解剖分布可能相当于半影区
Objective To investigate the change and role of heat shock protein 70 (HSP70) after focal cerebral ischemia / reperfusion in rats. Methods The middle cerebral artery occlusion (MCAO) model of ischemia - reperfusion was established by thread plug method. Immunohistochemistry was used to observe the histological distribution of HSP70. The relative content of HSP70 mRNA in ischemic cortex and basal ganglia was detected by RT-PCR. Results There were fewer HSP70 positive cells in basal ganglia, shorter duration, more HSP70 positive cells in cortex and longer duration. RT-PCR results showed that the relative content of HSP70 m RNA consistent with the results of immunohistochemistry. Conclusion HSP70 is a sensitive indicator of neuronal damage in ischemic area. The distribution of HSP70 in ischemic cortex may be equivalent to that of the penumbra
