目的探讨抗独特型抗体(Ab2)2H4和5D3模拟抗原诱导免疫应答的能力。方法用Ab2免疫Balb/c小鼠获得抗血清 ,以ELISA检测其Ab3,WesternBlot分析其识别抗原的分子质量。用迟发型超敏反应(DTH)和淋巴细胞增殖试验 ,检测Ab2诱发细胞免疫反应的能力。结果2H4,5D3免疫同系动物产生的含有抗抗独特型抗体(Ab3)的抗血清 ,可特异性地与靶细胞结合。FC2(Ab1)和经2H4免疫的Ab3可识别相对分子质量(Mr)相同约68000的抗原 ,且不同于HNL5(Ab1)和5D3免疫的Ab3所识别的抗原(Mr80000)。在DTH试验中 ,所致小鼠足垫肿大的程度 ,2H4和5D3实验组均显著高于对照组。而在淋巴细胞增殖试验中 ,Ab2免疫鼠脾T细胞在体外对靶细胞或Ab2的再次刺激呈现明显的细胞增殖反应 ,而无关肿瘤细胞或抗体的刺激则不能引起细胞增殖。结论抗独特型抗体2H4和5D3具有鼻咽癌相关抗原的内影像 ,能模拟不同分子质量的鼻咽癌相关抗原诱导体液和细胞免疫应答。
Objective To investigate the ability of anti-idiotypic antibodies (Ab2) 2H4 and 5D3 mimic antigens to induce immune responses. Methods Balb / c mice were immunized with Ab2 to obtain antiserum. Ab3 was detected by ELISA. The molecular weight of antigen was identified by Western Blot. Using delayed-type hypersensitivity (DTH) and lymphocyte proliferation assays, the ability of Ab2 to induce cellular immune responses was examined. Results Antiserum containing anti-idiotypic antibody (Ab3) produced by 2H4 and 5D3-immunocompetent animals could specifically bind to target cells. FC2 (Ab1) and Ab3 immunized with 2H4 recognized an antigen with the same molecular weight (Mr) of about 68,000 and different from the antigen recognized by Ab3 (Mr80000) immunized with HNL5 (Ab1) and 5D3. In the DTH test, the degree of footpad swelling was significantly higher in the 2H4 and 5D3 experimental groups than in the control group. In lymphocyte proliferation assay, spleen T cells immunized with Ab2 showed significant cell proliferative response to target cells or Ab2 re-stimulation in vitro, while unrelated tumor cells or antibody stimulation did not induce cell proliferation. Conclusion Anti-idiotypic antibodies 2H4 and 5D3 have the internal images of nasopharyngeal carcinoma-associated antigens, which can simulate the humoral and cellular immune responses induced by different molecular weights of NPC-associated antigens.
