目的:观察肝宁颗粒剂对四氯化碳(CCl4)所致大鼠急性肝损伤的保护作用。方法:将Wistar大鼠随机分为正常组、模型组、肝宁颗粒剂高、中、低剂量组、醇提制剂组和联苯双酯组,ig给药10 d。各组于实验第5,9 d,sc CCl4造模,第10 d,摘眼球取血,分离血清,以血清肝功能、SOD的活力及MDA含量以及肝脏组织病理学的改变为观察指标。结果:与模型组比较,肝宁颗粒剂高、中剂量组、醇提制剂组、联苯双酯组的血清ALT、AST、ALP、MDA均显著下降(P<0.01或P<0.05),血清SOD显著升高(P<0.01或P<0.05);肝脏组织学检查表明,肝宁颗粒剂能明显减轻肝细胞变性、坏死以及炎细胞浸润。肝宁颗粒剂各项测定指标的作用强度均不如联苯双酯。结论:肝宁颗粒剂对CCl4所致大鼠急性肝损伤有明显的降酶保肝作用。
Objective: To observe the protective effect of Ganning Granules on acute liver injury induced by carbon tetrachloride (CCl4) in rats. METHODS: Wistar rats were randomly divided into normal group, model group, high-, medium-, low-dose group of Ganning granules, ethanol extraction group and bifendate group, and ig administration for 10 days. In each group, on the 5th and 9th day of the experiment, scCCl4 was modeled. On the 10th day, blood was collected from the eyeball and serum was separated. The serum liver function, SOD activity and MDA content, and histopathological changes in the liver were taken as the observation indexes. Results:Compared with the model group, the serum ALT, AST, ALP, and MDA levels in the high, middle dose, alcohol extract, and bifendate groups of Ganning Granules significantly decreased (P<0.01 or P<0.05). SOD was significantly increased (P<0.01 or P<0.05); liver histological examination showed that Ganning Granules can significantly reduce liver cell degeneration, necrosis, and inflammatory cell infiltration. The strength of the various indicators of Ganning granules are not as strong as bifendate. Conclusion: Ganning granules have obvious protective effects on hepatic injury induced by CCl4 in rats.
