目的 :研究cAMP、cGMP在心肌细胞碱性成纤维细胞生长因子 (bFGF)基因表达调节中的作用。方法 :分离培养大鼠心肌细胞 ,加入 8-溴 -cAMP和硝普钠 ,升高细胞内cAMP、cGMP浓度 ,用原位杂交及免疫细胞化学的方法观察细胞内bFGF的表达 ,图象分析检测bFGF表达量的变化。结果 :在培养中加 8-溴 -cAMP的心肌细胞 ,2 4h时bFGFmRNA及蛋白表达均明显高于对照组 ,48h、72h仍然较强表达 (P <0 0 5 )。而加硝普钠的心肌细胞 ,bFGF表达明显低于对照组 ,以 48h最为显著 (P <0 0 1)。结论 :cAMP、cGMP可能与心肌细胞bFGF的表达调节有关
Objective: To investigate the role of cAMP and cGMP in the regulation of basic fibroblast growth factor (bFGF) gene expression in cardiomyocytes. Methods: Myocardial cells were isolated and cultured, and 8-bromo-cAMP and sodium nitroprusside were added to increase the intracellular cAMP and cGMP concentrations. The expression of bFGF in cells was observed by in situ hybridization and immunocytochemistry. bFGF expression changes. Results: After cultured with 8-bromo-cAMP cardiomyocytes, the expression of bFGF mRNA and protein were significantly higher at 24 hours and still remained at 48 hours and 72 hours (P <0.05). However, the expression of bFGF in cardiomyocytes with sodium nitroprusside was significantly lower than that in the control group (48h) (P <0.01). Conclusion: cAMP and cGMP may be involved in the regulation of bFGF expression in cardiomyocytes
