右归胶囊对肾阳虚大鼠血管紧张素系统影响的实验研究

目的本研究立足肺肾相关理论,以肌注氢化可的松建立肾阳虚大鼠模型,并给予右归胶囊进行干预,以血管紧张素系统AngⅡ、AT1R指标探讨温补肾养法及肾阳虚对“肺朝百脉”的影响。方法健康雄性SD大鼠60只,随机分为对照组、模型组、干预组,对照组不造模,生理盐水灌胃;模型组造模,生理盐水灌胃;干预组造模,右归胶囊混悬液灌胃。取大鼠肺组织Western blot法检测AngⅡ、AT1R的的表达。结果模型组合干预组大鼠第16天时,AngⅡ和AT1R明显少于正常组,差异有统计学意义(P<0.05);第46天时,与正常组比较,模型组大鼠肺组织AngⅡ表达减少,AT1R表达也减少,差异有统计学意义(P<0.05);与模型组比较,干预组大鼠肺组织AngⅡ和AT1R表达较多,差异有统计学意义(P<0.05),干预组第46天AngⅡ明显增高,与16天干预组相比,差异有统计学意义(P<0.05)。结论肾阳虚大鼠体内血液循环相关因子AngⅡ、AT1R受到影响,右归胶囊可以通过增加模型大鼠肺组织AngⅡ和AT1R的表达,从而缓解肾阳虚对“肺朝百脉”的影响。 Objective This study based on the theory of lung and kidney, intramuscular injection of hydrocortisone to establish the rat model of kidney yang, and give the right to the capsule for intervention, with angiotensin system Ang Ⅱ, AT1R indicators of warming and kidney yang and kidney yang Impact on the “lung toward the hundred”. Methods Sixty healthy male Sprague-Dawley rats were randomly divided into control group, model group, intervention group and control group. Rats in control group were intragastrically infused with normal saline. The model group and saline were given intragastrically. Suspension gavage. The expression of AngⅡ and AT1R in rat lung tissue was detected by Western blot. Results On the 16th day, the AngⅡ and AT1R in the model group were significantly lower than those in the normal group (P <0.05). On the 46th day, compared with the normal group, the expression of Ang Ⅱ in the lung tissue of the model group decreased, (P <0.05). Compared with the model group, the expression of AngⅡ and AT1R in the lung tissue of the intervention group was more than that of the model group (P <0.05), and the expression of AT1R was also decreased Compared with 16-day intervention group, the difference was statistically significant (P <0.05). Conclusion AngⅡ and AT1R of blood circulation related factors in Kidney Yang deficiency rats are affected. Yougui Capsule can relieve the effects of Kidney Yang deficiency on “Lung towards Lotus” by increasing the expression of Ang Ⅱ and AT1R in the lungs of model rats .
