目的:探讨温肾醒脑方对VD模型大鼠学习记忆功能、神经干细胞增殖及b FGF基因表达的影响,阐明温肾醒脑方益智醒脑开窍的机理。方法:采用“劳倦过度、房室不节”法、高脂饮食法以及反复性脑缺血再灌注结合降压法复制肾(阳)虚痰瘀型VD模型,将动物随机分为正常组、假手术组、模型组、温肾醒脑方组(中药组),于造模后1 d、30 d处死动物,评价动物的认知功能,免疫组织化学法观察nestin阳性细胞,ELISA和RT-PCR检测b FGF表达水平。结果:造模后动物逃避潜伏期明显延长,中药治疗组与模型组相比较,逃避潜伏期时间明显缩短差异具有统计学意义(P<0.05);中药组与假手术组相比,差异无显著性(P>0.05)。正常大鼠海马和皮层有少量nestin阳性细胞,造模后假手术组、模型组和中药组大鼠nestin阳性细胞数稍有增加,差异无统计学意义(P>0.05);30天后模型组、假手术组与正常组nestin阳性细胞数基本相近,差异无统计学意义(P>0.05),中药治疗组在海马与皮层均有显著增加,与正常组、模型组相比差异具有统计学意义(P<0.05)。正常组脑组织有低水平b FGF蛋白和基因表达,造模后模型组、假手术组与中药治疗组表达增强,中药治疗组、模型组与正常组相比差异有统计学意义(P<0.05),但两组间差异无统计学意义(P>0.05);中药治疗组b FGF水平维持在比较高的水平,而模型组基本恢复到正常组水平,中药治疗组与模型组、正常组相比差异均有统计学意义(P<0.05)。结论:温肾醒脑方能促进认知功能的恢复,增强VD大鼠神经干细胞增殖和b FGF表达,是其益智醒脑开窍的可能机理。
Objective: To investigate the effects of Wenshen Xingnao Recipe on learning and memory function, neural stem cell proliferation and b FGF gene expression in VD rats, and to elucidate the mechanism of Wenshen Xingnao Fangzhi Zhi Xingnao resuscitation. Methods: Kidney (Yang) deficiency phlegm and blood stasis type VD model was duplicated with “overwork, atrioventricular node” method, high fat diet method and repeated cerebral ischemia / reperfusion combined with antihypertensive method. Animals were randomly divided into normal Group, sham operation group, model group and Wenshen Xingnao recipe group (Chinese medicine group). Animals were sacrificed on the 1st and 30th day after modeling to evaluate the cognitive function of the animals. Immunohistochemistry was used to observe the expression of nestin positive cells, ELISA and The expression of b FGF was detected by RT-PCR. Results: After modeling, the evasion latency of animals was significantly prolonged. Compared with the model group, the evasion latency was significantly shortened (P <0.05). There was no significant difference between the traditional Chinese medicine group and the sham operation group P> 0.05). There was a small amount of nestin positive cells in the hippocampus and cortex of normal rats. There was a slight increase in the number of nestin positive cells in sham operation group, model group and TCM group after modeling (P> 0.05) The number of nestin positive cells in sham-operation group and normal group were almost the same, with no significant difference (P> 0.05). There was a significant increase in hippocampus and cortex in Chinese medicine group compared with normal group and model group (P <0.05) P <0.05). The normal group had low level of b-FGF protein and gene expression, and the expression of the model group, sham-operation group and TCM-treated group was enhanced after the model was established. There was significant difference between the traditional Chinese medicine treatment group and the model group and the normal group (P <0.05 ), But there was no significant difference between the two groups (P> 0.05). The level of b FGF in traditional Chinese medicine group was maintained at a relatively high level, while the model group returned to the level of normal group. The differences were statistically significant (P <0.05). Conclusion: Wenshen Xingnao Decoction can promote the recovery of cognitive function, enhance the proliferation of neural stem cells and the expression of b FGF in VD rats, which is the possible mechanism of recuperation.
