目的:观察脂多糖及一些前炎性细胞因子对体外培养的星形胶质细胞产生一氧化氮(NO)的影响。方法:Griess法检测细胞培养上清中NO_2-含量。结果:体外培养4h时人BT325星形胶质瘤细胞开始产生NO,12h达高峰(15.0-17.5μmol·L~(-1))并持续至72h.LPS 1mg·L~(-1),IFN-γ100kU·L~(-1),TNF-α100kU·L~(-1),IL-1 100kU·L~(-1)及IL-2100kU·L~(-1)可增强体外培养的BT325细胞产生NO,以TNF-α,IL-1和IL-2作用较为明显,四种细胞因子合用则作用更强。结论:炎症刺激或前炎性细胞因子促进神经胶质细胞产生NO。
Objective: To observe the effect of lipopolysaccharide and some pro-inflammatory cytokines on the production of nitric oxide (NO) in cultured astrocytes. Methods: Griess method was used to detect NO 2 - content in cell culture supernatant. RESULTS: The human BT325 astroglioma cells started to produce NO at 4h and reached the peak at 12h (15.0-17.5μmol·L -1) for 72h.LPS 1mg · L -1, IFN (100kU · L -1, IL-1 100kU · L -1 and IL-2100kU · L -1) enhanced the proliferation of BT325 cells in vitro Produce NO, the role of TNF-α, IL-1 and IL-2 is more obvious, the combination of the four cytokines is more effective. Conclusion: Inflammatory stimuli or proinflammatory cytokines promote NO production in glial cells.
