目的探讨氟化钠(NaF)对人成骨肉瘤MG-63细胞增殖的影响。方法以0、10-2、10-1、1、10、102、5×102、103、5×103、104、2×104、4×104μmol/L NaF染毒MG-63细胞,培养48 h。采用MTT实验观察NaF对MG-63细胞增殖能力的影响,以流式细胞技术检测MG-63细胞周期变化。结果 MTT实验结果显示,MG-63细胞开始有增殖活性时的NaF浓度为102μmol/L,增殖活性最大时的浓度为5×103μmol/L,增殖活性受到抑制时的浓度为2×104μmol/L。流式细胞技术检测结果显示,采用MTT实验中确定的3个关键浓度染毒细胞48 h后,高剂量(2×104μmol/L)组的MG-63细胞凋亡率最高(6.001%),细胞增殖指数(PI)最低(0.339);中剂量(5×103μmol/L)组的细胞凋亡率最低(2.220%),PI最高(0.632);低剂量(102μmol/L)组的PI及细胞凋亡率均与对照组无明显差异。结论 NaF对成骨细胞具有双向作用,低浓度下促进细胞增殖,超过一定浓度后因其增殖抑制而使细胞凋亡增加。
Objective To investigate the effect of sodium fluoride (NaF) on the proliferation of human osteosarcoma MG-63 cells. Methods MG-63 cells were treated with 0,10-2,10-1,1,10,102,5 × 102,103,5 × 103,104,2 × 104,4 × 104μmol / L NaF for 48 h . The effect of NaF on the proliferation of MG-63 cells was observed by MTT assay. The cell cycle of MG-63 cells was detected by flow cytometry. Results The results of MTT assay showed that the concentration of NaF was 102μmol / L, the proliferation rate was 5 × 103μmol / L and the inhibitory concentration was 2 × 104μmol / L when MG-63 cells started to proliferate. The results of flow cytometry showed that MG-63 cells in high dose (2 × 104μmol / L) group had the highest apoptotic rate (6.001%) after being exposed to 3 key concentrations of MTT for 48 h, (PI) was the lowest (0.339); the apoptosis rate was the lowest (2.220%) and the highest PI (0.632) at medium dosage (5 × 103μmol / L) Mortality were no significant difference with the control group. Conclusion NaF has a bidirectional effect on osteoblasts and promotes cell proliferation at low concentrations. After a certain concentration of NaF, NaF increases apoptosis due to its inhibition of proliferation.
