目的构建针对血小板源性血管内皮生长因子(PDECGF)小干扰RNA(siRNA)表达质粒和阴性对照质粒pIASRC-PDECGF,并观察对PDECGF表达的影响。方法构建针对PDECGF siRNA表达质粒pIASR-PDECGF和阴性对照质粒pIASRC-PDECGF,将沟建成功的质粒转染大肠癌细胞株GC7901,观察PDECGF蛋白表达的影响。结果成功构建针对PDECGF siRNA表达质粒pIASR-PDECGF,并发现它能够明显抑制PDECGF的表达。而阴性对照质粒则无此作用。结论血小板源性内皮细胞生长因子小干扰RNA能够特异性抑制PDECGF的表达。
Objective To construct a small interfering RNA (siRNA) expression plasmid for platelet-derived vascular endothelial growth factor (PDECGF) and a negative control plasmid pIASRC-PDECGF, and observe the effect on the expression of PDECGF. METHODS: The PDECGF siRNA expression plasmid pIASR-PDECGF and the negative control plasmid pIASRC-PDECGF were constructed. The plasmids were successfully transfected into the colorectal cancer cell line GC7901 and the effect of PDECGF protein expression was observed. Results The recombinant plasmid pIASR-PDECGF targeting PDECGF was successfully constructed and it was found that it significantly inhibited the expression of PDECGF. The negative control plasmid does not have this effect. Conclusion The platelet-derived endothelial growth factor siRNA can specifically inhibit the expression of PDECGF.
