目的:研究干扰素α对肝癌细胞中转录因子Sp1(specificity protein 1)的表达和活性的影响。方法:采用蛋白免疫印迹法分析干扰素α对肝癌细胞株MHCC97H中Sp1的表达及磷酸化水平的影响。采用凝胶电泳迁移率变动分析法检测干扰素α对Sp1活性的影响。结果:干扰素α可显著下调Sp1的表达和磷酸化水平,以对磷酸化水平的下调更为明显;干扰素α可显著抑制Sp1的活性。结论:Sp1可能作为干扰素α抗肝癌的临床疗效的预测指标。
Objective: To investigate the effect of interferon α on the expression and activity of transcription factor Sp1 in hepatocellular carcinoma cells. Methods: The effect of interferon α on Sp1 expression and phosphorylation in hepatocellular carcinoma cell line MHCC97H was analyzed by Western blotting. The effect of interferon alpha on Sp1 activity was examined by gel electrophoretic mobility shift assay. Results: Interferon α significantly down-regulated the expression and phosphorylation of Sp1, which was more pronounced down-regulation of phosphorylation. Interferon α significantly inhibited the activity of Sp1. Conclusion: Sp1 may serve as a predictor of the clinical efficacy of interferon α against liver cancer.
