目的 :研究柯萨奇B病毒 (CVB)感染对 3种人胰岛 β细胞和正常人胰岛 β细胞 (NBC)表面CD5 6表达的影响。方法 :利用流式细胞仪技术分析病毒作用 3种人胰岛 β细胞CM、HP6 2、TRM6以及NBC前后细胞表面表达CD5 6的变化。结果 :病毒作用前不同人胰岛 β细胞CD5 6表达量不同 ,HP6 2细胞表达量最高 ,其次是NBC ,CM细胞表达量最低。CVB3和CVB4作用于CM细胞 2 4h促进细胞表面CD5 6的表达 ,作用 4 8h细胞表面CD5 6的表达量下降。CVB3和CVB4作用于HP6 2细胞、TRM6细胞、NBC 2 4h、4 8h细胞表面CD5 6的表达量降低。结论 :随着病毒作用人胰岛 β细胞时间的延长 ,细胞表面CD5 6表达量下降 ,CD5 6是否在IDDM致病中起作用有待进一步研究。
Objective: To investigate the effect of Coxsackie B virus (CVB) infection on the expression of CD56 on the surface of three kinds of human islet β cells and normal human islet β cells (NBC). Methods: Flow cytometry was used to analyze the changes of cell surface expression of CD5 6 on three kinds of human pancreatic β cells CM, HP6 2, TRM6 and NBC before and after virus infection. Results: The expression of CD5 6 in different human pancreatic β-cells was different before virus infection. The expression of CD56 was the highest in HP6 2 cells, followed by NBC and the lowest in CM cells. The effect of CVB3 and CVB4 on CM cells for 24 h promoted the expression of CD5 6 on the cell surface, and decreased the expression of CD5 6 on the 48 h. CVB3 and CVB4 on HP6 2 cells, TRM6 cells, NBC 2 4h, 48h cell surface CD5 6 expression decreased. CONCLUSION: With the prolongation of viral β-cell in human islets, the expression of CD5 6 on the cell surface is decreased, and whether CD5 6 plays a role in the pathogenesis of IDDM remains to be further studied.
