目的探讨吸入式甲醛暴露对小鼠骨髓组织中转录因子GATA-2和CCAAT/增强子结合蛋白(C/EBPα)在mRNA以及蛋白表达水平的影响。方法将24只Balb/C小鼠随机分为3组,每组8只,采用仿真式口鼻暴露的方式,暴露于不同浓度的气态甲醛环境中(0、0.5和3.0 mg/m3),每天8 h,连续2周。染毒结束后,分别采用反转录PCR(RT-PCR)和酶联免疫吸附(ELISA)法测定小鼠骨髓组织中转录因子GATA-2和C/EBPαmRNA和蛋白的表达。结果与对照组相比,转录因子GATA-2和C/EBPαmRNA和蛋白的表达均随甲醛浓度的升高呈下降趋势,且甲醛浓度为3.0 mg/m3时,差异有统计学意义(P<0.05)。结论高浓度(3.0 mg/m3)甲醛暴露可以抑制小鼠骨髓组织中GATA-2和C/EBPαmRNA和蛋白的正常表达。
Objective To investigate the effect of inhaled formaldehyde exposure on mRNA and protein expression of transcription factors GATA-2 and CCAAT / enhancer binding protein (C / EBPα) in mouse bone marrow. Methods Twenty-four Balb / C mice were randomly divided into 3 groups (8 rats in each group). The animals were exposed to different concentrations of gaseous formaldehyde (0, 0.5 and 3.0 mg / m3) 8 h for 2 weeks. At the end of exposure, mRNA and protein expressions of GATA-2 and C / EBPα in bone marrow of mice were determined by reverse transcription-polymerase chain reaction (RT-PCR) and enzyme-linked immunosorbent assay (ELISA) Results Compared with the control group, the expression of GATA-2 and C / EBPα mRNA and protein of the transcription factors decreased with the increase of formaldehyde concentration, and the difference was statistically significant at the formaldehyde concentration of 3.0 mg / m3 (P <0.05 ). Conclusions High concentration (3.0 mg / m3) formaldehyde exposure can inhibit the normal expression of GATA-2 and C / EBPα mRNA and protein in mouse bone marrow.
