目的在原代培养的新生大鼠心肌细胞上,观察三七总皂苷(PNS)对血管紧张素Ⅱ(AngⅡ)诱导的心肌细胞肥大的影响。方法应用细胞培养技术培养新生大鼠心肌细胞,采用相差显微镜测量细胞大小,测定心肌细胞3H-亮氨酸掺入作为心肌细胞肥大的指标;用流式细胞仪测定心肌细胞凋亡率,用Westernblot法检测心肌细胞促凋亡蛋白Bax和抑制凋亡蛋白Bcl-2的表达。结果 PNS能抑制AngⅡ诱导的心肌细胞增大、心肌细胞3H-亮氨酸掺入率的显著增加(P<0.05)、心肌细胞凋亡率的增加(P<0.05)、心肌细胞促凋亡蛋白Bax的表达明显增高(P<0.05),心肌细胞抑制凋亡蛋白Bcl-2的表达明显降低(P<0.05),其效果与AngⅡ受体阻滞剂缬沙坦相似。结论 PNS可以抑制AngⅡ诱导的心肌细胞肥大,这可能与其同时抑制了心肌细胞的凋亡有关。
Objective To observe the effect of Panax notoginseng saponins (PNS) on cardiomyocyte hypertrophy induced by angiotensin II (Ang II) in primary cultured neonatal rat cardiomyocytes. Methods Cell culture was used to culture neonatal rat cardiomyocytes. The size of the cells was measured by phase contrast microscopy. The incorporation of 3H-leucine in cardiomyocytes was used as an indicator of myocardial hypertrophy. The apoptosis rate of cardiomyocytes was measured by flow cytometry. Western blotting was used. Methods The expression of pro-apoptosis protein Bax and inhibition of apoptosis protein Bcl-2 in cardiomyocytes was detected. Results PNS inhibited the increase of cardiomyocytes induced by AngII, significantly increased the incorporation rate of 3H-leucine in cardiomyocytes (P<0.05), increased the apoptosis rate of cardiomyocytes (P<0.05), and promoted apoptosis of cardiomyocytes. The expression of Bax was significantly increased (P<0.05), and the expression of apoptosis protein Bcl-2 was significantly decreased in cardiomyocytes (P<0.05). The effect was similar to that of valsartan, an AngII receptor blocker. Conclusion PNS can inhibit the hypertrophy of cardiomyocytes induced by AngII, which may be related to the inhibition of myocardial cell apoptosis at the same time.
