目的研究2型糖尿病患者脂肪组织内胰岛素受体后信号传导分子蛋白酪氨酸磷酸酶1B(protein tyrosine phosphatase 1B,PTP1B)和含2个SH2结构的蛋白酪氨酸磷酸酶(protein tyrosine phosphatase with two src-homology 2,SH-PTP2)的蛋白表达,探讨人体脂肪组织产生胰岛素抵抗的分子机制。方法用Western blot技术检测来自民航总医院及北京大学第一医院外科住院,并接受腹部手术治疗的患者[根据口服葡萄糖耐量试验的结果将其分为2型糖尿病组(n=10)和非糖尿病对照组(n=10)]的皮下及其大网膜脂肪组织内PTP1B、SH-PTP2的蛋白表达水平。结果 PTP1B的蛋白表达(IA)在2型糖尿病患者皮下(24.14±0.67)及其大网膜脂肪组织内(28.35±1.12)均较对照组增加[皮下14.53±0.52(P=0.042);大网膜18.65±2.84(P=0.035)];SH-PTP2在其皮下(42.14±6.67)及大网膜脂肪组织内(41.88±11.57)的蛋白表达虽然亦较对照组(皮下35.34±8.21;大网膜29.98±6.38)增加,但差异无统计学意义。结论 2型糖尿病患者脂肪组织内PTP1B及SH-PTP2蛋白表达的异常改变,可能是引起其产生胰岛素抵抗的分子机制之一。
Objective To investigate the expression of insulin tyrosine phosphatase 1B (PTP1B) and two tyrosine phosphatases with two tyrosine phosphatases (SHP) in type 2 diabetic patients. src-homology 2, SH-PTP2) in human adipose tissue to explore the molecular mechanism of insulin resistance in human adipose tissue. Methods Western blot was used to detect patients hospitalized from the Civil Aviation General Hospital and the First Hospital of Peking University undergoing surgical treatment and undergoing abdominal surgery. They were divided into type 2 diabetes (n = 10) and non-diabetes according to oral glucose tolerance test Control group (n = 10)] subcutaneous and omental adipose tissue PTP1B, SH-PTP2 protein expression levels. Results The expression of PTP1B protein in subcutaneous (24.14 ± 0.67) and omentum adipose tissue (28.35 ± 1.12) in type 2 diabetic patients was significantly higher than that in control group [subcutaneous 14.53 ± 0.52 (P = 0.042; Membrane 18.65 ± 2.84 (P = 0.035)]. The protein expression of SH-PTP2 in subcutaneously (42.14 ± 6.67) and omental adipose tissue (41.88 ± 11.57) was significantly higher than that in control group (35.34 ± 8.21, Membrane 29.98 ± 6.38) increased, but the difference was not statistically significant. Conclusions Abnormal changes of PTP1B and SH-PTP2 protein expression in adipose tissue of type 2 diabetic patients may be one of the molecular mechanisms that cause insulin resistance.
