目的:探讨白细胞介素6(IL-6)对宫颈癌C33a细胞侵袭能力的影响。方法:宫颈癌C33a细胞分为对照组和siIL-6组(RNA干扰处理组),利用新型阳离子脂质体转染试剂Hilymax导入IL-6 RNA干扰序列,并检测IL-6基因的表达,而后应用Transwell实验检测C33a细胞侵袭能力的改变。结果:与对照组比较,qPCR实验结果显示,RNA干扰组IL-6 mRNA下降了41%(P<0.01),显示成功干扰了IL-6的基因表达;Transwell实验结果显示,较之于对照组,干扰组IL-6 C33a细胞侵袭细胞数减少(P<0.01)。结论:本研究成功干扰了IL-6基因表达,并且IL-6表达抑制导致C33a细胞侵袭能力下降。
Objective: To investigate the effect of interleukin-6 (IL-6) on invasiveness of cervical carcinoma C33a cells. Methods: Cervical cancer C33a cells were divided into control group and siIL-6 group (RNA interference treatment group). IL-6 RNA interference sequence was introduced into Hilymax by a novel cationic liposome transfection reagent and the expression of IL-6 gene was detected. Transwell assay was used to detect the invasion ability of C33a cells. Results: Compared with the control group, the results of qPCR showed that IL-6 mRNA decreased by 41% (P <0.01) in the RNA interference group, which showed that the gene expression of IL-6 was successfully disrupted. The results of Transwell assay showed that compared with the control group , And the number of invasive cells in IL-6 C33a cells decreased in the interference group (P <0.01). Conclusion: This study successfully interfered with IL-6 gene expression, and inhibition of IL-6 expression led to decreased invasion ability of C33a cells.
