补肾和补肾健脾方对尾部悬吊大鼠股骨骨丢失的作用研究

目的:观察补肾和补肾健脾方对尾部悬吊大鼠股骨的影响,比较它们防护骨丢失作用机制的异同。方法:采用头低位-30°尾部悬吊模拟失重性骨丢失,实验结束后,ELISA法测血清碱性磷酸酶(ALP)活性、胰岛素样生长因子(IGF-1)和骨桥蛋白(OPN)含量,计算右侧股骨矿化沉积速率,观察左侧股骨骨形态计量学静态参数。结果:与正常组比较,悬吊组大鼠血清ALP活性、IGF-1和OPN含量、矿化沉积速率、骨小梁面积比、骨小梁厚度和骨小梁数量均显著降低(P<0.01),骨小梁分离度明显增高(P<0.01);与悬吊组比较,补肾组血清ALP活性、血清IGF-1含量、矿化沉积速率、骨小梁面积比、骨小梁厚度和骨小梁数量升高(P<0.01,P<0.05),骨小梁分离度明显降低(P<0.01);补肾健脾组大鼠血清ALP活性、IGF-1含量、OPN含量、矿化沉积速率、骨小梁面积比、骨小梁厚度和骨小梁数量均显著升高(P<0.01,P<0.05),骨小梁分离度明显降低(P<0.01);较之补肾组,补肾健脾组大鼠血清ALP活性和矿化沉积速率升高(P<0.05),骨小梁分离度降低(P<0.05)。结论:补肾和补肾健脾方均对模拟失重引起的股骨骨丢失具有防护作用,以补肾健脾方作用为优,其作用机制可能与提高成骨细胞活性、促进骨形成有关。 Objective: To observe the effect of Bushen and Bushen Jianpi Recipe on the femur of tail-suspended rats, and to compare their mechanism of bone loss. Methods: The weight loss bone loss was simulated by tail suspension at -30 °. After the experiment, serum alkaline phosphatase (ALP) activity, insulin-like growth factor (IGF-1) and osteopontin (OPN) Content, calculate the right femoral mineralization deposition rate, observation left femoral bone morphometry metrology static parameters. Results: Compared with normal group, serum ALP activity, IGF-1 and OPN contents, mineralization deposition rate, trabecular area ratio, trabecular thickness and trabecular number in suspension group were significantly decreased (P <0.01 ), The trabecular separation was significantly higher (P <0.01). Compared with the suspension group, serum ALP activity, serum IGF-1 level, mineralization deposition rate, trabecular area ratio, trabecular thickness and bone (P <0.01, P <0.05), and the trabecular separation was significantly lower (P <0.01). The serum ALP activity, IGF-1 content, OPN content, mineralization deposition rate , Trabecular area ratio, trabecular thickness and trabecular number were significantly increased (P <0.01, P <0.05), and trabecular separation was significantly decreased (P <0.01). Compared with Bushen group, Bushenjian The serum ALP activity and mineralization deposition rate in spleen group were increased (P <0.05), and the trabecular separation was decreased (P <0.05). Conclusion: Bushen and Bushen Jianpi Recipe have the protective effect on the loss of femur caused by simulated weightlessness. The effect of Bushen Jianpi Recipe is superior to that of Bushen Jianpi Recipe. The mechanism may be related to improving osteoblast activity and promoting bone formation.
