目的 探讨细胞周期素依赖激酶 (CDKs)抑制因子p2 7表达对神经母细胞瘤 (NB)发生发展的影响。方法 检测 2 6例NB标本中p2 7基因表达 ,结合增殖细胞核抗原 (PCNA)表达和细胞凋亡检测结果进行分析。结果 III及IVS期、神经节母细胞瘤 (GNB)、预后好组织 (FH )型标本p2 7表达指数 (EI)为 2 7.2 8± 13 .61、3 4 .60± 12 .16、2 6.2 9± 14 .71,显著高于Ⅲ~Ⅳ期、NB、预后差组织 (UFH )型标本 15 .81± 12 .92、18.77± 14 .77、13 .3 4± 13 .79(P值均 <0 .0 5 ) ;Ⅰ~Ⅱ或IVS期、GNB、FH型标本PCNAEI为 9.47± 8.43、0 .83± 0 .5 7、11.0 5± 10 .71则显著低于Ⅲ~Ⅳ期、NB、UFH型标本 2 7.0 5± 19.2 4、2 4.44± 17.42、3 2 .0 9± 17.0 4(P值均 <0 .0 5 )。p2 7EI与有丝分裂核碎裂指数 (MKI)呈负相关 (n =2 6,r =-0 .5 13 ) ,与NB细胞凋亡指数呈正相关 (n =17,r =0 .490 ) ,与存活时间呈正相关 (n =12 ,r =0 .63 5 ) ;PCNAEI与MKI呈正相关 (n =2 0 ,r =0 .90 3 )。结论 p2 7抑制细胞增殖周期及诱导细胞凋亡的功能 ,在NB的发病机制中可能具有重要作用
Objective To investigate the effect of cyclin-dependent kinase (CDKs) inhibitor p27 expression on the development of neuroblastoma (NB). Methods The p27 gene expression in NB specimens was detected in 26 patients. The results were analyzed by combining the expression of proliferating cell nuclear antigen (PCNA) and apoptosis. Results The expression index (EI) of p2 7 in stage III and IVS, ganglioalganglionoma (GNB), and good-prognosis tissue (FH) samples was 27.28±13.61, 34.60±12.16, and 26.2. 9±14.71, significantly higher than those of III-IV, NB, and poorly-differentiated tissue (UFH) specimens, 15.81±12.92, 18.77±14.77, and 13.43±13.79 (P <0. 0 5 ); PCNAEI of I-II or IVS, GNB and FH specimens was 9.47±8.43, 0.83±0.57, and 11.05±10.71, which were significantly lower than those of III-IV and NB. The UFH type specimens were 2 7.0 5± 19.2 4, 2 4.44± 17.42, 3 2 .0 9± 17.0 4 (P values were both <0. 0 5). P2 7EI was negatively correlated with the mitotic nuclear fragmentation index (MKI) (n =26,r = -0.53) and was positively correlated with the apoptotic index of NB cells (n =17, r = .490). There was a positive correlation between survival time (n =12, r = 0.635); PCNAEI was positively correlated with MKI (n = 20, r = 0.93). Conclusion The function of p27 inhibiting cell proliferation cycle and inducing cell apoptosis may play an important role in the pathogenesis of NB
