目的:研究莲子壳中的黄酮类化学成分及抗氧化活性。方法:莲子壳的60%乙醇提取物经过大孔树脂初步分离之后,采用Toyopearl HW-40S凝胶柱进一步分离得到了6个组分(LF-F1~LF-F6),再采用高效液相以及液质联用系统对其中的组分2(LF-F2)组成及结构进行分析,并采用化学发光法等方法研究了LF-F2的抗氧化活性。结果:莲子壳黄酮粗提物中总黄酮为94.1%;LF-F2中黄酮甙元是由槲皮素、杨梅黄酮、山奈酚、异鼠李素等组成的。LF-F2对超氧阴离子自由基、羟自由基以及对双氧水的清除作用的IC50分别为420、40、0.6μg/ml。结论:从莲子壳中鉴定出LF-F2中有芦丁存在。LF-F2具有很好的抗氧化效果,并存在剂量效应关系。
Objective: To study the flavonoids and antioxidant activity in lotus seed hull. METHODS: After 60% ethanol extract of lotus seed shell was separated by macroporous resin, six components (LF-F1-LF-F6) were further separated by using Toyopearl HW-40S gel column, and then the high-performance liquid phase was used. The composition and structure of component 2 (LF-F2) were analyzed by LC/MS system, and the anti-oxidation activity of LF-F2 was studied by chemiluminescence and other methods. Results: The total flavonoids in the crude extract of flavonoids from lotus seed hull were 94.1%. The flavonoids in LF-F2 were composed of quercetin, myricetin, kaempferol and isorhamnetin. The IC50 values of LF-F2 for scavenging superoxide anion radicals, hydroxyl radicals, and hydrogen peroxide were 420, 40, and 0.6 μg/ml, respectively. Conclusion: The presence of rutin in LF-F2 was identified from lotus seeds. LF-F2 has a good antioxidant effect, and there is a dose-effect relationship.
