目的:优化HPD-600型大孔吸附树脂分离纯化肿节风总黄酮的工艺条件和参数。方法:以肿节风总黄酮含量及回收率等为指标,考察肿节风总黄酮的大孔吸附树脂分离纯化工艺效果及影响因素。结果:最佳纯化工艺为上样液pH 5.0,总黄酮质量浓度15~20 g.L-1,上样液和树脂比10∶1,3 BV水洗,3 BV70%乙醇洗脱,得总黄酮纯度大于80%的提取物,总黄酮转移率大于85%。结论:HPD-600型大孔吸附树脂对肿节风总黄酮分离纯化效果较好,适合工业化大生产。
OBJECTIVE: To optimize the conditions and parameters for the separation and purification of total flavonoids from Arthritis phaeozem by HPD-600 macroporous adsorption resin. Methods: According to the total flavonoids content and recovery rate of swollen-sectioned wind, the effect and influencing factors of separation and purification of total-flavone macroporous adsorption resin were studied. Results: The optimal purification process was pH5.0, total flavonoids concentration of 15 ~ 20 gL-1, the sample and resin ratio of 10: 1, 3 BV washing, 3 BV70% ethanol, the total flavonoids purity greater than 80% of the extract, total flavonoid transfer rate greater than 85%. Conclusion: HPD-600 macroporous resin has good effect on the separation and purification of total flavonoids from Arthritis tumefaciens and is suitable for industrialized large-scale production.
