目的探讨~99Tc-亚甲基二膦酸盐(~99Tc-MDP)对胶原诱导性关节炎(collagen induced arthritis,CIA)大鼠的关节炎症、骨质破坏的治疗作用及其对肿瘤坏死因子(TNF)-α的影响。方法以大鼠CIA为动物模型,予以~99Tc-MDP对其进行干预,通过观察关节肿胀变化、放射学和组织病理学及细胞因子TNF-α水平变化评价药物的治疗作用结果大体评分显示:~99Tc-MDP对关节肿胀具有明显抑制作用。放射学评分和组织学切片显示:~99Tc-MDP能明显减轻骨质破坏。~99Tc-MDP治疗组血清TNF-α水平明显低于模型对照组。结论~99Tc-MDP可减轻CIA炎症和抑制骨质破坏;并能显著抑制TNF-α的产生。
Objective To investigate the therapeutic effect of 99Tc-methylene diphosphonate (~ 99Tc-MDP) on joint inflammation and bone destruction in collagen induced arthritis (CIA) rats and its effect on tumor necrosis factor TNF) -α. Methods Rat CIA was used as an animal model and treated with ~ 99Tc-MDP. The therapeutic effect of the drug was evaluated by observing changes of joint swelling, radiology, histopathology and cytokines TNF-α. The results showed that ~ 99Tc-MDP has a significant inhibitory effect on joint swelling. Radiological scores and histological sections showed: ~ 99Tc-MDP can significantly reduce bone destruction. ~ 99Tc-MDP treatment group serum TNF-α levels were significantly lower than the model control group. Conclusion ~ 99Tc-MDP can reduce the inflammation of CIA and inhibit bone destruction; and can significantly inhibit the production of TNF-α.
