目的:建立高效液相色谱法测定金辛镇痛喷雾剂中氢溴酸东莨菪碱和硫酸阿托品的含量。方法:采用Agilent ZorbaxC_(18)(250mm×4.6mm,5μm)色谱柱,流动相为乙腈-0.07mol·L~(-1)磷酸钠溶液(含0.40%三乙胺,17.50mmol·L~(-1)十二烷基硫酸钠,用磷酸调pH至6.00)(42:58),流速:0.8ml·min~(-1),检测波长:215nm,柱温:40℃。结果:氢溴酸东莨菪碱在1.00~5.00μg范围内线性关系良好,r=0.9999(n=5),平均回收率98.35%,RSD=1.68%(n=6);硫酸阿托品在2.94~14.70μg范围内呈良好的线性,r=0.9997(n=6),平均回收率为97.71%,RSD=1.91%(n=6)。结论:本法简便、准确、灵敏度高,重复性好。
Objective: To establish a HPLC method for the determination of scopolamine hydrobromide and atropine sulfate in Jinxin analgesic spray. Methods: Agilent Zorbax C_ (18) (250 mm × 4.6 mm, 5 μm) column was used. The mobile phase consisted of acetonitrile-0.07 mol·L -1 sodium phosphate solution containing 0.40% triethylamine and 17.50 mmol·L ~ -1) sodium dodecyl sulfate adjusted to pH 6.00 with phosphoric acid (42:58), flow rate: 0.8 ml · min -1, detection wavelength: 215 nm, column temperature: 40 ℃. Results: Scopolamine hydrobromide had a good linear relationship in the range of 1.00-5.00 μg with r = 0.9999 (n = 5), the average recovery was 98.35% and RSD was 1.68% (n = 6). Atropine sulfate ranged from 2.94 to 14.70 μg R = 0.9997 (n = 6). The average recovery was 97.71%, RSD = 1.91% (n = 6). Conclusion: This method is simple, accurate, sensitive and reproducible.
