目的建立糖尿病(DM)、DM前期患者血浆微小RNA(miRNA)差异表达谱,寻找DM及DM前期相关的miRNA。方法利用miRNA芯片技术,筛查健康人、DM患者和DM前期患者血浆中差异表达的miRNA;采用生物信息学方法预测差异miRNA的靶基因并分析其生物学功能。结果比较三者血浆,基因芯片检测共发现4个差异表达的miRNA(P<0.05),生物信息学技术预测了4个miRNA的靶基因,主要与多细胞有机体的发展、信号转导、细胞分化、凋亡、离子转运、细胞代谢过程调控等生物学功能相关。结论获得的DM和DM前期患者血浆中存在差异表达的miRNA,可能是DM早期诊断新的生物标志物和潜在的治疗靶点。
Objective To establish differential expression profiles of plasma microRNAs (miRNAs) in patients with diabetes mellitus (DM) and pre-DM and to find pre-DM miRNAs. Methods The miRNAs differentially expressed in the plasma of healthy people, DM patients and pre-DM patients were screened by miRNA microarray. Bioinformatics methods were used to predict the target genes of miRNAs and their biological functions were analyzed. Results There were four miRNAs differentially expressed in plasma and gene chip (P <0.05). Bioinformatics techniques predicted the target genes of four miRNAs, which were mainly related to the development of multicellular organisms, signal transduction and cell differentiation , Apoptosis, ion transport, regulation of cellular metabolism and other biological functions. Conclusions There are differentially expressed miRNAs in the plasma of patients with pre-DM and DM, which may be new biomarkers for DM early diagnosis and potential therapeutic targets.
