目的:探讨参芪复方对GK大鼠2型糖尿病大血管病变的内皮保护作用与机制。方法:5月龄雄性SPF级GK大鼠按血糖水平随机分为GK空白组、模型组、西药阿托伐他汀组、中药参芪复方组,另设正常Wistar对照组。模型、西药、中药组给予L-NAME0.1mg·mL-1·d-1,加入饮用水中进行造模。Wistar组喂饲普通饲料,其余各组喂饲高脂饲料时间为35天。造模同时开始给予相应受试药物,周期35天。实验结束时,腹主动脉采血,冰上取主动脉。HE染色对腹主动脉进行形态学观察,双抗体夹心ABC-ELISA法测定主动脉VEGF含量,实时荧光定量PCR技术检测主动脉ICAM-1mRNA的表达水平,western blot法检测大鼠主动脉ICAM-1蛋白表达。结果:参芪复方能显著改善GK大鼠腹主动脉形态学变化。参芪复方组大鼠的主动脉VEGF含量、主动脉ICAM-1mRNA及ICAM-1蛋白表达较模型组均显著降低(P<0.05)。结论:降低主动脉VEGF含量,抑制主动脉ICAM-1mRNA及蛋白表达可能是参芪复方保护血管内皮功能、防治DM大血管病变的部分作用机制。
Objective: To investigate the protective effects and mechanism of Shenqi Compound on type 2 diabetic macroangiopathy in GK rats. Methods: Five-month-old male SPF-GK rats were randomly divided into GK group, model group, atorvastatin group, Shenqi Compound group and normal Wistar control group. Model, western medicine, Chinese medicine group given L-NAME0.1mg · mL-1 · d-1, added to drinking water modeling. Wistar group fed normal feed, the remaining groups fed high-fat feed time is 35 days. Modeling at the same time began to give the appropriate test drugs, cycle 35 days. At the end of the experiment, blood was collected from the abdominal aorta and the aorta was taken on ice. The morphology of the abdominal aorta was observed by HE staining. The content of VEGF in the aorta was detected by double antibody sandwich ABC-ELISA. ICAM-1 mRNA expression was detected by real-time fluorescence quantitative PCR. ICAM-1 Protein. Results: Shenqi Compound can significantly improve the morphological changes of abdominal aorta in GK rats. Compared with the model group, the content of VEGF and the expression of ICAM-1mRNA and ICAM-1 in the aorta of Shenqi Compound group were significantly decreased (P <0.05). CONCLUSION: Reducing the content of VEGF in the aorta and inhibiting the expression of ICAM-1 mRNA and protein in the aorta may be part of the mechanism of Shenqi compound in protecting vascular endothelial function and preventing and treating DM macroangiopathy.
