门冬氨酸鸟氨酸与2,4-二硝基氟苯在碱性条件下衍生化后,以C18柱分离,采用HPLC法测定门冬氨酸鸟氨酸注射液中的门冬氨酸鸟氨酸;以0.3 mol·L-1醋酸盐缓冲液-甲醇(40∶60)为流动相,流速1.0 mL·min-1,检测波长360 nm,空白不干扰门冬氨酸鸟氨酸的测定;0.4~1.6 mg·mL-1门冬氨酸鸟氨酸与其峰面积的线性关系良好(r=0.9999),其含量的RSD<2.0%(n=12),测定溶液在12 h内稳定,高、中、低浓度下的平均回收率为100.02%,RSD=0.97%。所用方法专属性强、线性关系好、溶液稳定,准确度及精密度好,能够满足门冬氨酸鸟氨酸注射液中门冬氨酸鸟氨酸的含量测定。
Aspartate ornithine was derivatized with 2,4-dinitrofluorobenzene under basic conditions and was separated on a C18 column. The concentration of ornithine ornithine in 0.4-1.6 mg · mL -1 ascorbic acid had a good linear relationship with its peak area The average recoveries at steady, high, medium and low concentrations were 100.02% with RSD = 0.97%. The method used has the advantages of strong specificity, good linearity, stable solution, good accuracy and precision, and can meet the determination of aspartate ornithine in aspartate ornithine injection.
