目的 :研究 3种细胞因子 (肿瘤坏死因子 α ,干扰素 γ和IL 6 )对乳腺癌细胞钠碘转运体 (NIS)基因表达的影响 ,探讨乳腺肿瘤组织NIS的表达特点和调控因素。方法 :采用乳腺癌细胞株MCF 7进行培养 ,分别给予不同浓度的肿瘤坏死因子 α、干扰素 γ或IL 6进行刺激 72h培养。采用RT PCR方法检测细胞中NIS的mRNA表达情况。结果 :乳腺癌细胞MCF7在经过肿瘤坏死因子 α、干扰素 γ或IL 6刺激后 ,细胞中NIS的mRNA表达比对照组有明显降低 ,并且与细胞因子浓度浓度成反比趋势。结论 :细胞因子肿瘤坏死因子 α、干扰素 γ和IL 6对乳腺癌细胞MCF 7的NIS基因表达有抑制作用 ,这提示细胞因子可能通过调节乳腺癌NIS基因的表达 ,从而影响乳腺癌细胞对放射性碘治疗的敏感性。
Objective: To investigate the effects of three cytokines (tumor necrosis factor alpha, interferon gamma and IL 6) on the expression of sodium iodide symporter (NIS) in breast cancer cells and to explore the expression characteristics and regulation of NIS in breast tumor. Methods: Breast cancer cell line MCF 7 was used for culture, and different concentrations of tumor necrosis factor α, interferon γ or IL 6 were respectively cultured for 72 hours. RT-PCR method was used to detect the mRNA expression of NIS in cells. Results: The mRNA expression of NIS in breast cancer cell MCF7 was significantly lower than that in the control group after being stimulated by tumor necrosis factor α, interferon γ or IL 6, and was inversely proportional to the concentration of cytokines. CONCLUSION: TNF-α, IFN-γ and IL-6 inhibit the NIS gene expression in breast cancer cell line MCF-7, which suggests that cytokines may influence the expression of NIS gene in breast cancer cells, Iodine treatment sensitivity.
