采用15%PEG6000竖直滤纸法对41份陆地棉种子进行萌发期抗旱性研究,分别测定在胁迫条件下及清水对照条件下的发芽势、发芽率、3d芽长3、d芽重、7d下胚轴长、7d胚根长、胚根长/胚芽长、芽长生长率等8个指标。结果表明,这8个指标在PEG6000胁迫条件下,均比对照降低,除3d芽重外,41份材料的变异系数均大大增加,说明陆地棉种子萌发受15%(-0.4MPa)PEG6000的抑制,且材料间受抑制程度差异较大。同时计算出胁迫条件下与对照条件的相对值,通过对这8个指标的相对值进行方差分析和主成分分析,筛选出相对发芽势、相对发芽率、相对7d下胚轴长、相对7d胚根长、相对胚根长/胚芽长、芽长生长率等7个指标与陆地棉萌发期抗旱性有关。利用这7个指标对41份陆地棉材料进行隶属函数分析和相关性分析,并以平均隶属函数值代表抗旱性强弱,平均隶属函数值越大,抗旱性越强。结果表明,相对7d胚根长与平均隶属函数值存在着极显著的正相关(r=0.839),同时筛选出陆地棉萌发期抗旱性较强的材料7份:冀668、鲁棉研21、9409选系、DP99B、创棉22、sGK中980、邯177;抗旱性中等的材料有10份:中23A-12、中23A抗棉F12、中棉所35、冀1286、GKZ19F6、双豫97-2067、光籽2、耐高温-2、中S9612、光籽1。
The drought resistance of 41 cultivated G. hirsutum seeds at germination stage was studied by vertical filter paper method with 15% PEG6000. The germination potential, germination rate, 3 d bud length, 3 d bud weight and 7 d under stress and water control conditions were respectively measured. The length of embryo axis, the length of embryo of 7 days, the length of radicle of embryo, the length of embryo and length of bud and the growth of bud. The results showed that all the eight indexes were lower than those of the control under PEG6000 stress. By relative analysis of variance and principal component analysis, the relative germination energy, relative germination rate, relative 7d hypocotyl length, Seven indexes, root length, relative radicle length / bud length and bud length growth rate, were related to the drought resistance of upland cotton. Using these seven indexes, the membership function analysis and correlation analysis of 41 Upland cotton materials were carried out. The mean membership function value was used to represent the drought resistance. The larger the average membership function, the stronger the drought resistance. The results showed that there was a significant positive correlation (r = 0.839) between the radicle length and the average membership function at 7d, and 7 materials with stronger drought resistance at the germination stage were also screened: Ji 668, Lumianyan 21, 9409 selected line, DP99B, a cotton 22, sGK 980, Hand 177; medium drought resistance materials are 10: 23A-12, 23A anti-cotton F12, cotton 35, Hebei 1286, GKZ19F6, Shuangyu 97 -2067, Light Seed 2, High Temperature-2, Medium S9612, Light Seed 1.
