目的研究猴耳环Pithecellobium clypearia Benth枝叶的化学成分及其免疫活性。方法采用多种色谱学方法进行分离纯化,利用波谱学方法进行结构鉴定;并通过ConA诱导的T淋巴细胞增殖试验考察各化合物的免疫活性。结果从中分离并鉴定了8个化合物,分别为(-)-表没食子儿茶素(1),(-)-5,7,3’,4’,5’-五羟基黄烷(2),(-)-表没食子儿茶素-7-没食子酸酯(3),(-)-5,3’,4’,5’-四羟基黄烷-7-没食子酸酯(4),槲皮素-3-O-α-L-吡喃鼠李糖苷(5),杨梅树皮素-3-O-α-L-吡喃鼠李糖苷(6),没食子酸(7),没食子酸乙酯(8)。结论化合物3和8为首次从该属植物中分离得到,化合物1为首次从该种植物中分离得到;化合物3能显著抑制ConA诱导的T淋巴细胞增殖,其IC50为4.4μmol·Lˉ1。
Objective To study the chemical composition and immunological activity of the leaves of Pithecellobium clypearia Benth. Methods A variety of chromatographic methods were used to separate and purify. The structures were identified by spectroscopy. The immune activity of each compound was examined by Con A-induced T lymphocyte proliferation assay. Results Eight compounds were isolated and identified from (-)-epigallocatechin (1), (-)-5,7,3’,4’,5’-pentahydroxyflavan (2). (-)-Epigallocatechin-7-gallate (3), (-)-5,3’,4’,5’-tetrahydroxyflavan-7-gallate (4), suede -3-O-α-L-pyrhamnoside (5), Myricitin-3-O-α-L-rhamnopyranoside (6), Gallic acid (7), Gallic acid B Ester (8). Conclusion Compounds 3 and 8 were isolated from this genus for the first time. Compound 1 was isolated from the plant for the first time. Compound 3 significantly inhibited Con A-induced proliferation of T lymphocytes with an IC50 of 4.4 μmol·Lˉ1.
