目的探讨一种高效、简便和稳定的羊水细胞染色体制备方法,提高产前诊断的准确性和成功率。方法选择2002年9月3日到2009年7月9日孕周为16.0~29的产前筛查为高风险的孕妇2689位,抽取羊水进行细胞培养,行羊水细胞染色体核型分析。结果 2689例羊水细胞培养成功率为99.9%,核型分析的成功率为99.8%,发现异常核型74例,异常核型率为2.8%,其中18-三体10例,占0.37%,21-三体19例,占0.71%。结论本方法具有高效、简便、染色体带纹清晰等优点,在产前诊断中具有结果准确可靠和实用价值。
OBJECTIVE: To study an efficient, simple and stable method for the preparation of amniotic fluid cell chromosomes to improve the accuracy and success rate of prenatal diagnosis. Methods 2689 pregnant women with high risk of prenatal screening from September 3, 2002 to July 9, 2009 were enrolled. A total of 2689 pregnant women with high risk of prenatal screening were enrolled in this study. Amniotic fluid was used for cell culture and amniotic fluid cell karyotype analysis. Results The success rate of 2689 cases of amniotic fluid cell culture was 99.9%, and the success rate of karyotype analysis was 99.8%. There were 74 cases of abnormal karyotype with abnormal karyotype rate of 2.8%, including 18 cases of trisomy 18, accounting for 0.37%, 21 - trisomy in 19 cases, accounting for 0.71%. Conclusion The method has the advantages of high efficiency, simpleness, clear chromosomes, and so on. It has accurate, reliable and practical results in prenatal diagnosis.
