目的通过运用蛋白质组学技术寻找舌癌血清特异性标记物,为舌癌早期诊断、病程监测以及预后判断提供可靠的血清学指标。方法采用舌癌 Tca8113细胞系体外培养,并移植于裸鼠。以正常裸鼠血清为对照,采用双向凝胶电泳分离技术分析荷瘤小鼠的血清蛋白表达谱,并通过质谱鉴定差异表达蛋白。结果在荷瘤裸鼠血清中分离和鉴定了5个差异表达蛋白,其中鳞状细胞癌抗原1检索得分最高,为97分,定量分析中变化最大,并与舌癌密切相关。结论荷瘤裸鼠血清中鳞状细胞癌抗原1高表达,有望成为舌癌的特异性血清标记物之一。
Objective To search for serum specific markers of tongue cancer by using proteomics technology and provide reliable serological markers for tongue cancer diagnosis, course monitoring and prognosis. Methods The tongue cancer Tca8113 cell line was cultured in vitro and transplanted in nude mice. Serum samples of the tumor-bearing mice were analyzed by two-dimensional gel electrophoresis using normal nude mice as control. The differentially expressed proteins were identified by mass spectrometry. Results Five differentially expressed proteins were isolated and identified from the serum of nude mice bearing tumor. The highest score of SCC - 1 was found in SCC - 1, with a maximum of 97 and the most significant change in quantitative analysis, which was closely related to tongue cancer. Conclusion High expression of squamous cell carcinoma antigen 1 in the serum of nude mice is expected to become one of the specific serum markers for tongue cancer.
