目的:观察肝型脂肪酸结合蛋白(L-FABP)在大鼠非酒精性脂肪肝(NAFLD)模型中的动态表达,探讨L-FABP在大鼠NAFLD所起的作用。方法:选取Wistar大鼠40只,随机分为对照组和模型组。其中对照组给予普通饲料,模型组给予高脂饲料。分别于4,8,12周处死。测血清中的丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、血清胆固醇(TC)、血清甘油三酯(TG)。并测定空腹血糖(FBG)、空腹胰岛素(FINS),计算ISI。HE染色观察肝脏脂肪组织病理变化。并应用半定量反转录聚合酶链反应(RT-PCR)方法测定大鼠的肝脏组织中L-FABPmRNA的表达。结果:4周、8周1、2周模型组大鼠肝脏中L-FABPmRNA表达增强(P<0.01),且随着造模时间的延长,L-FABP的表达逐渐增高,与对照组差异显著(P<0.01)。模型组血清ALT、AST、TC、TG的水平均较对照组显著增高(P<0.01),而ISI明显降低(P<0.01)。结论:持续12周的高脂饮食可以成功复制大鼠NAFLD模型,且随着造模时间的延长可引起L-FABP表达逐渐增强。
Objective: To observe the dynamic expression of hepatic fatty acid binding protein (L-FABP) in rat non-alcoholic fatty liver (NAFLD) model and to explore the role of L-FABP in rat NAFLD. Methods: Forty Wistar rats were randomly divided into control group and model group. The control group was given normal feed, the model group was given high-fat feed. Were killed at 4,8,12 weeks. Serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), serum cholesterol (TC), serum triglyceride (TG). Fasting blood glucose (FBG) and fasting insulin (FINS) were measured to calculate ISI. The pathological changes of liver adipose tissue were observed by HE staining. The expression of L-FABP mRNA in the liver of rats was detected by semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR). Results: The expression of L-FABP mRNA in the liver of model group was significantly increased at 4 weeks, 8 weeks and 8 weeks (P <0.01), and the expression of L-FABP increased gradually with the extension of modeling time, which was significantly different from that of control group (P <0.01). The levels of serum ALT, AST, TC and TG in the model group were significantly higher than those in the control group (P <0.01), but the ISI was significantly lower (P <0.01). CONCLUSION: A high-fat diet lasting 12 weeks can successfully replicate rat NAFLD model, and the expression of L-FABP can be gradually increased with the prolongation of modeling time.
