目的:通过检测溃克灵对溃疡性结肠炎模型大鼠血清白细胞介素-8(IL-8)含量及肠黏膜一氧化氮(NO)含量、一氧化氮合酶(NOS)活性的影响,探讨溃克灵防治溃疡性结肠炎的作用机理。方法:采用TNBS法(100mg/kgTNBS+50%的乙醇0.25mL)灌肠复制出溃疡性结肠炎大鼠模型。将72只大鼠随机分为正常组、模型组、SASP(柳氮磺吡啶)组、溃克灵大、中、小剂量组(每组12只)共6组,分别对各组大鼠血清IL-8含量及肠黏膜NO含量、NOS活性进行检测。结果:与正常组比较,模型组大鼠血清IL-8含量显著升高(P<0.01),肠黏膜NO含量、NOS活性显著升高(P<0.01);与模型组比较,SASP组、溃克灵大剂量组可显著降低IL-8含量(P<0.01),降低结肠黏膜NO含量、NOS活性(P<0.01)。结论:溃克灵对UC大鼠有治疗作用,降低IL-8含量,降低结肠黏膜NO含量,NOS活性可能是其治疗机制之一。
OBJECTIVE: To detect the effect of Kuikeling on the levels of serum interleukin-8 (IL-8), nitric oxide (NO) and nitric oxide synthase (NOS) in rat model of ulcerative colitis, To investigate the mechanism of ulkaline in the prevention and treatment of ulcerative colitis. Methods: The rat model of ulcerative colitis was replicated by TNBS method (100mg / kg TNBS + 50% ethanol 0.25mL). 72 rats were randomly divided into normal group, model group, SASP (sulfasalazine) group, uracil large, medium and small dose groups (12 in each group) a total of 6 groups, respectively, the serum of rats in each group IL-8 content and intestinal mucosal NO content, NOS activity were detected. Results: Compared with the normal group, the content of IL-8 in the model group was significantly increased (P <0.01), and the content of NO and the activity of NOS in the intestinal mucosa were significantly increased (P <0.01). Compared with the model group, Ketamine high dose group can significantly reduce the content of IL-8 (P <0.01), reduce the colon mucosal NO content, NOS activity (P <0.01). Conclusion: Kuikuling has the therapeutic effect on UC rats, reducing the content of IL-8 and decreasing the content of NO in colonic mucosa. NOS activity may be one of its therapeutic mechanisms.
