目的比较化橘红6种黄酮成分抑制血小板聚集的活性,并对其构效关系进行分析。方法分别制备大鼠富血小板血浆(PRP)和洗涤血小板,采用比浊法检测6种黄酮成分在PRP或洗涤血小板中对二磷酸腺苷(ADP)诱导的大鼠血小板聚集的抑制作用。结果与对照组比较,6种黄酮成分均能显著抑制ADP诱导的大鼠血小板聚集,其中川陈皮素抑制作用最强,随后依次是桔红素、橙皮素、柚皮素或橙皮苷、柚皮苷;与在PRP抑制血小板聚集活性比较,黄酮成分在洗涤血小板中抑制血小板聚集的活性显著提高,并且其抗血小板的构效关系与在PRP中是一致的。结论化橘红黄酮对大鼠PRP或洗涤血小板聚集均有明显抑制作用,其抑制血小板聚集的活性与其结构密切相关,甲氧基和酚羟基数目以及A环C7羟基可能影响黄酮抑制血小板聚集的生物活性。
Objective To compare the inhibitory effect of 6 flavonoids of orange juice on the platelet aggregation activity, and to analyze its structure-activity relationship. Methods Platelet-rich plasma (PRP) and platelet-washing were prepared respectively. The inhibitory effect of 6 flavonoids on platelet aggregation induced by adenosine diphosphate (ADP) in PRP or washed platelets was detected by turbidimetry. Results Compared with the control group, all the six flavonoids could significantly inhibit platelet aggregation induced by ADP, of which, nobiletin had the strongest inhibitory effect, followed by tannin, hesperetin, naringenin or hesperidin, Naringin. Compared with the inhibition of platelet aggregation by PRP, the activity of flavonoids in inhibiting platelet aggregation in washing platelets was significantly increased, and its anti-platelet effect was consistent with that in PRP. CONCLUSION: The orange-yellow flavonoids have a significant inhibitory effect on PRP or platelet aggregation in rats, and its inhibitory effect on platelet aggregation is closely related to its structure. The number of methoxy and phenolic hydroxyl groups and the C7 hydroxyl of A ring may affect the biological activity of flavonoids in inhibiting platelet aggregation .
