大孔吸附树脂的不同预处理对贝母多糖纯化效果影响

目的:筛选出最佳的大孔吸附树脂预处理方法,以提高太白贝母粗多糖大孔吸附树脂的纯化效果。方法:AB-8型大孔吸附树脂经过已报道的4种不同预处理方法处理后,比较其对太白贝母粗多糖核酸脱除率、蛋白质脱除率、色素脱除率的影响,以及对多糖损失率和羟基自由基清除效果的影响。结果:对于太白贝母粗多糖,经过预处理方法1处理过的大孔吸附树脂对粗多糖纯化效果最佳,其核酸清除率为58.55%,蛋白质清除率为61.73%。420 nm和470 nm检测波长处色素脱除率在46.14%~64.65%,多糖损失率为43.01%,多糖羟基自由基清除率为25.07%。对于甘孜川贝母粗多糖,经过预处理方法3处理的大孔吸附树脂对粗多糖纯化效果最佳,预处理方法1次之。结论:预处理方法1对于2种贝母多糖溶液的纯化效果均较好。 OBJECTIVE: To select the best pretreatment method for macroporous adsorption resin to improve the purification effect of macroporous adsorption resin. Methods: After the AB-8 macroporous resin was treated by four different pretreatment methods reported, the effect of removal rate of nucleic acid, protein removal rate, Polysaccharide loss rate and hydroxyl radical scavenging effect. Results: For the crude polysaccharide from Taibai Fritillaria, the macroporous resin treated with pretreatment method 1 had the best purification effect on crude polysaccharides with a nucleic acid clearance rate of 58.55% and a protein clearance rate of 61.73%. The detection rates of pigment removal at 420 nm and 470 nm ranged from 46.14% to 64.65%, the polysaccharide loss rate was 43.01% and the hydroxyl radical scavenging rate of polysaccharide was 25.07%. For Ganzi Fritillary crude polysaccharides, macroporous resin treated by pretreatment method 3 had the best purification effect on crude polysaccharides, and the pretreatment method was the first one. Conclusion: The pretreatment method 1 for the two Fritillaria polysaccharide solution were better purification.
