吸烟与温石棉单独及联合作用对人胚肺细胞内[Ca~(2+)]i的影响

目的 研究香烟烟雾溶液 (CSS)与温石棉混悬液 (CH)单独及联合作用对人胚肺 (HEL)细胞内游离钙离子浓度 ([Ca2 +]i)的影响。方法 体外培养的HEL细胞用Fluo 3/AM标记细胞内Ca2 +,用不同浓度CSS和CH作用 1h ,荧光分光光度计测定细胞内 [Ca2 +]i。结果  1.2 5× 10 -4 、2 .5 0× 10 -4 、5 .0 0× 10 -4 支 /mlCSS单独作用于HEL细胞 1h ,可使 [Ca2 +]i分别升高 5 0 .4%、75 .4%和10 9.3% ;2 0、40、80 μg/mlCH单独作用于HEL细胞 1h ,可使[Ca2 +]i分别升高 3 .0 %、5 3.1%和 116 .7%。CSS及CH单独作用均具有明显的剂量 -效应关系 (前者r =0 .96 3 ,P =0 .0 37;后者r =0 .976 ,P =0 .0 2 4) ;当 1.2 5× 10 -4 支 /mlCSS与 40 μg/mlCH联合作用、2 .5 0× 10 -4 支 /mlCSS与 2 0 μg/mlCH联合作用时均可协同增加 [Ca2 +]i。结论 CSS与CH可在诱导细胞信号转导方面发挥协同作用 Objective To investigate the effects of cigarette smoke solution (CSS) and chrysotile (CH) suspension alone or in combination on the intracellular free calcium concentration ([Ca2 +] i) in human embryo lung cells. Methods HEL cells cultured in vitro were labeled with Fluo 3 / AM intracellular Ca2 +, with different concentrations of CSS and CH for 1 h, intracellular [Ca2 +] i was measured by fluorescence spectrophotometer. Results After treated with 5 × 10 -4, 2.5 × 10 -4, 5.0 × 10 -4 / ml CSF for 1 h, the [Ca 2+] i increased by 50.4% , 75.4%, and 10 9.3%, respectively. When 20, 40, 80 μg / ml CHCH alone acted on HEL cells for 1h, [Ca2 +] i increased by 3.0%, 5.31% and 116.7%, respectively. There was a significant dose-effect relationship between CSS and CH alone (former r = 0.96 3, P = 0. 037; latter r = 0. 976, P = 0. 0224) 10 -4 / mlCSS combined with 40 μg / ml CH, 2.50 × 10 -4 / mlCSS combined with 20 μg / ml CH synergistically increased [Ca 2+] i. Conclusions CSS and CH can play synergistic roles in inducing cell signal transduction
