目的 观察日本血吸虫单克隆抗独特型抗体NP30主动免疫小鼠诱导的保护性免疫力。方法 选择L16 (4× 2 12 )正交设计 ,8周龄昆明种小鼠随机分为 16个实验组和 2个对照组。对照组腹腔接种SP2 / 0腹水。所有小鼠在腹部皮肤感染 10 0± 2条日本血吸虫尾蚴 ,尾蚴攻击后第 30天处死小鼠 ,行左心室 -门静脉灌注收集成虫 ,计算减虫率。结果 根据不同的免疫方案 ,NP30主动免疫可产生 2 2 36 % - 5 0 4 6 %的减虫率 ,并确定了最优免疫方案。结论 提示NP30主动免疫对尾蚴攻击可产生一定的保护力 ,具有血吸虫疫苗候选分子的潜能。
Objective To observe the protective immunity induced by the active immunization of mice with the monoclonal anti-idiotypic antibody NP30 of Schistosoma japonicum. Methods L16 (4 × 2 12) orthogonal design was used. Kunming mice of 8 weeks old were randomly divided into 16 experimental groups and 2 control groups. The control group was intraperitoneally inoculated with SP2 / 0 ascites. All mice were infected with 100 ± 2 S. japonicum cercariae on the skin of the abdomen. The mice were killed on the 30th day after cercariae challenge. The adults were collected by left ventricular-portal vein perfusion to calculate the worm reduction rate. Results According to the different immunization protocols, NP30 active immunization resulted in a reduction rate of 2 2 36% - 50 4 6% and the optimal immunization schedule was determined. Conclusions NP30 active immune vaccine cercariae attack can produce some protection, with the potential of schistosomiasis vaccine candidate molecules.
