目的:研究侧柏叶炮制前后槲皮素含量的变化,建立侧柏叶合理的炮制方法。方法:通过HPLC法对侧柏叶中槲皮素的含量进行测定,色谱柱为Agilent Eclipse XPB-C18(4.6 mm×250 mm,5μm);流动相为甲醇:0.2%磷酸溶液(55∶45);流速1.0 mL·min-1;检测波长360 nm;柱温30℃;并对侧柏叶的生品、烘制品及不同炮制品中槲皮素进行测定和比较。结果:槲皮素在0.03~0.52μg成良好的线性关系,回收率为97.5%~100.8%,RSD1.22%,槲皮素在烘制品中的含量较传统炮制品高。结论:所建立的HPLC法简便可靠、精密度高。烘制法具有和传统炮制法相接近的结果,提示其作为侧柏叶的炮制法存在一定的可行性。
Objective: To study the changes of quercetin content before and after the processing of oriental arborvitae leaves, and to establish the rational processing method of oriental arborvitae leaves. Methods: The content of quercetin in oriental arborvitae was determined by HPLC. The column was Agilent Eclipse XPB-C18 (4.6 mm × 250 mm, 5 μm). The mobile phase consisted of methanol: 0.2% phosphoric acid solution (55:45) ; The flow rate was 1.0 mL · min-1; the detection wavelength was 360 nm; the column temperature was 30 ℃; and the contents of quercetin in the unripe leaves, dried fruits and different processed products were measured and compared. Results: The calibration curve of quercetin was linear in the range of 0.03-0.52 μg. The recoveries were 97.5% -100.8% and the RSD was 1.22%. The content of quercetin in the dried products was higher than that of the traditional processed products. Conclusion: The established HPLC method is simple and reliable with high precision. Baking method with the traditional method of processing is close to the results, suggesting that the processing method as a Oriental Arborvitae there is a certain feasibility.
