目的 观察视网膜色素上皮 (retinal pigment epithelium,RPE)细胞白细胞介素 6 (inter-leukine 6 ,IL - 6 )和白细胞介素 8(interleukine 8,IL - 8)的表达及白细胞介素 1受体拮抗剂 (interleukine 1receptor antigonist,IL- 1ra)对 RPE细胞 IL- 6和 IL- 8表达的影响。 方法 培养的人 RPE细胞分为对照组和 IL - 1ra组 (IL - 1ra,1、10、10 0 ng/ ml) ,用酶联免疫吸附、免疫组织化学和原位杂交等方法观察 RPE细胞 IL- 6和 IL- 8的表达。 结果 对照组 RPE细胞培养上清液中 IL- 6、IL- 8分别为 2 0 0 0、5 0 0 0 pg/ ml。与对照组比较 ,IL - 1ra组 IL - 6、IL - 8分别为 30 0 pg/ ml(t=8.0 11,P <0 .0 1)、4 5 0 pg/ ml(t=11.4 4 6 ,P<0 .0 1)。经白细胞介素 1β(interleukine1β,IL- 1β)刺激后 RPE细胞 IL- 6和 IL- 8m RNA表达原位杂交表现为细胞浆中浓淡不一的蓝色着色 ,IL - 1ra作用组细胞染色较淡。 结论 在 IL - 1β刺激下 ,培养的人 RPE细胞可以表达 IL- 6、IL- 8的 m RNA和蛋白质 ,IL- 1ra能有效抑制其 IL- 6、IL- 8m RNA和蛋白质的表达
Objective To investigate the expression of interleukin 6 (IL - 6) and interleukin 8 (IL - 8) in retinal pigment epithelium (RPE) cells and the effects of interleukin 1 receptor antagonist (Interleukine 1 receptor antigonist, IL-1ra) on the expression of IL-6 and IL-8 in RPE cells. Methods Human RPE cells were divided into control group and IL - 1ra group (IL - 1ra, 1, 10 and 10 ng / ml). The levels of IL - - 6 and IL-8. Results The levels of IL-6 and IL-8 in the supernatant of RPE cells in the control group were respectively 20000 and 50000 pg / ml. Conclusions The human RPE cells can express m RNA and protein of IL - 6 and IL - 8 under the stimulation of IL - 1β, and IL - 1ra can effectively inhibit the expression of IL - 6, IL - 8m RNA and protein
