目的:证明非洛地平(Fel)对心肌钙通道是否有阻断作用,与硝苯啶和维拉帕米作一些比较.方法:利用细胞内微电极和单蔗糖电压钳方法研究Fel对豚鼠右心室乳头状肌动作电位,慢反应动作电位和慢钙电流的作用.结果:Fel呈浓度依赖性抑制动作电位的持续时间,慢反应动作电位的0相除极速度,幅度及持续时间和慢钙电流.Fel的作用不易冲洗掉,刺激频率增加,Fel作用减弱.Fel对心肌钙通道静息态有作用.用三氟拉嗪处理标本后,Fel作用消失.结果:Fel对心肌钙通道具有阻断作用.这种作用主要是对静息态钙通道的作用.
OBJECTIVE: To demonstrate whether Fel has a block effect on myocardial calcium channels, compared with nifedipine and verapamil. Methods: The intracellular microelectrode and monosuccinate voltage-clamp method were used to study the effects of Fel on the action potentials, slow response potentials and slow calcium currents in the right ventricular papillary muscles of guinea pigs. Results: The duration of action potentials, the 0 phase depolarization speed, the amplitude and duration of slow response action potentials and the slow calcium currents of Fel in a concentration - dependent manner. Fel’s role is not easy to wash out, stimulate the frequency increases, Fel weakened. Fel has a role in the resting state of cardiac calcium channels. Fel treated with trifluoperazine disappeared. Results: Fel had a blocking effect on myocardial calcium channels. This effect is mainly on the role of resting calcium channel.
