目的探讨冰片对低分化鼻咽癌(CNE-2Z)细胞氯通道的激活作用及对细胞容积的影响。方法采用全细胞膜片钳技术记录冰片激活CNE-2Z细胞膜电流,分析其电流特性;用活细胞图像法观察并测量冰片对细胞体积的影响。结果细胞外灌流冰片(20μmol·L-1)诱导CNE-2Z细胞膜氯通道开放,产生氯电流,该电流有明显外向优势,无电压及时间依赖性失活,其翻转电位接近氯离子平衡电位,该电流可被氯通道阻断剂tamoxifen抑制,细胞外灌流47%高渗溶液完全抑制该电流。细胞外灌流冰片30 min,细胞体积缩小约9.4%,该作用可被tamoxifen完全抑制。结论冰片可以激活低分化鼻咽癌细胞容积敏感性氯通道,导致细胞体积缩小,氯通道在冰片引起细胞体积缩小中起着重要作用。
Objective To investigate the effect of borneol on the activation of chloride channel and the cell volume in poorly differentiated nasopharyngeal carcinoma (CNE-2Z) cells. Methods Whole-cell patch-clamp technique was used to record the membrane current of CNE-2Z cells activated by borneol, and its current characteristics were analyzed. The effect of borneol on the cell volume was observed and measured by living cell imaging. Results Extracellular perfusion of borneol (20μmol·L-1) induced the opening of chloride channel in CNE-2Z cell membrane, resulting in a chloride current. The current had obvious extrinsic advantages and no voltage-dependent and time-dependent inactivation. This current can be inhibited by the chloride channel blocker tamoxifen, which is completely suppressed by extracellular infiltration of 47% hypertonic solution. The extracellular flux of borneol for 30 min reduced the cell volume by about 9.4%, which was completely inhibited by tamoxifen. CONCLUSION: Borneol can activate the volume-sensitive chloride channel in poorly differentiated nasopharyngeal carcinoma cell line, resulting in the decrease of cell volume. Chloride channel plays an important role in the reduction of cell volume caused by borneol.
