目的:通过动物模型实验观察益阴通络胶囊对局灶性脑缺血的影响,并探讨该药治疗缺血性脑血管病的可能机理。方法:围绕益阴通络胶囊的主要药效学研究,采用线栓法行右侧大脑中动脉阻塞造模,分组观察实验脑缺血大鼠脑组织半胱天冬蛋白酶~3(Caspase-3)的表达在假手术组、模型组中以及在益阴通络胶囊组和血塞通片对照药的作用下的不同变化,并进行统计分析。结果:采用线栓法行右侧大脑中动脉阻塞造模后,实验动物模型组大鼠脑组织中半胱天冬蛋白酶~3(Caspase-3)的含量显著增高,与假手术组相比有非常显著性差异(P<0.01)。益阴通络胶囊组Caspase-3较模型组明显减少,与模型组比较有非常显著性差异(P<0.01)。血塞通片对照组Caspase-3的含量较模型组无明显变化,无显著性差异(P>0.05)。结论:益阴通络胶囊对线栓法造成大鼠局灶性脑缺血具有很好的疗效,益阴通络胶囊可以下调局灶性脑缺血脑组织中Caspase-3的表达,间接或直接反应该药能清除自由基,降低炎性细胞因子及抑制病理产物,减轻脑水肿,减少神经元的损伤,从而抑制神经细胞调亡,保护脑细胞。
Objective: To observe the effect of Yiyin Tongluo Capsule on focal cerebral ischemia in animal models and to explore the possible mechanism of this medicine in the treatment of ischemic cerebrovascular disease. Methods: The main pharmacodynamic study of Yiyin Tongluo Capsule was made. The right middle cerebral artery was obstructed by the method of thread embolization. The changes of caspase-3 ) In sham-operation group, model group and Yiyintongluo Capsule group and Xuesaitong tablet control group, and analyzed statistically. Results: After blocking the right middle cerebral artery occlusion by thread occlusion, the content of Caspase-3 in brain tissue of experimental animal model group was significantly higher than that of sham-operated group Very significant difference (P <0.01). Caspase-3 in Yiyin Tongluo Capsule group was significantly lower than that in model group, which was significantly different from that in model group (P <0.01). Compared with the model group, the content of Caspase-3 in Xuesaitong tablet control group had no significant difference (P> 0.05). Conclusion: Yiyin Tongluo Capsule has a good curative effect on focal cerebral ischemia caused by thread plug method. Yiyin Tongluo Capsule can downregulate the expression of Caspase-3 in focal cerebral ischemia, Direct response to the drug can scavenge free radicals, reduce inflammatory cytokines and inhibit pathological products, reduce cerebral edema and reduce neuronal damage, thereby inhibiting the apoptosis of nerve cells, protecting the brain cells.
