目的研究槲皮素对大鼠原代肝细胞中I型血红素氧化酶(HO-1)蛋白表达及酶活性的影响。方法二步胶原酶技术分离培养大鼠原代肝细胞。用不同剂量(25、50、100、200 mol/L)槲皮素作用大鼠原代肝细胞12 h,或用50 mol/L槲皮素作用肝细胞不同时间(2~12 h)后,提取肝细胞微粒体,检测HO-1酶活性变化,并用WesternBlot方法检测HO-1蛋白表达水平的变化。结果大鼠原代肝细胞经50~200 mol/L槲皮素处理12 h,或用50 mol/L槲皮素处理4~12 h后,HO-1蛋白表达水平和酶活性与对照组比较明显升高(P<0.05)。结论槲皮素能够诱导大鼠原代肝细胞的HO-1蛋白表达,提高肝细胞内HO-1的酶活性。
Objective To investigate the effect of quercetin on the expression of type I heme oxygenase (HO-1) and the activity of enzymes in rat primary hepatocytes. Methods Two - step collagenase technique was used to isolate rat primary hepatocytes. After treated with quercetin at different doses (25, 50, 100, 200 mol / L) for 12 h, or treated with 50 mol / L quercetin for different time (2 ~ 12 h) The liver cell microsomes were extracted and the changes of HO-1 enzyme activity were detected. The changes of HO-1 protein expression were detected by Western blot. Results After primary hepatocytes were treated with 50 ~ 200 mol / L quercetin for 12 h or treated with 50 mol / L quercetin for 4 ~ 12 h, the expression of HO-1 protein and the activity of the enzyme were compared with those of the control group Was significantly higher (P <0.05). Conclusions Quercetin can induce HO-1 protein expression in rat primary hepatocytes and increase the activity of HO-1 in hepatocytes.
