本文通过用抗Collagen Ⅳ(Col.Ⅳ)抗体阻断Col.Ⅳ活性的方法,研究Col.Ⅳ失活的移植神经段(1cm)在植入大鼠坐骨神经后对再生轴突和非神经细胞的作用和影响.实验用90只Wistar大鼠.30只鼠双侧坐骨神经经退变处理后,切取1cm长的神经段作为供体神经.60只受体鼠随机分成对照组(30只)和抗(col.Ⅳ)组(30只).供体神经(60条)经5次冷冻和加热处理以杀死雪旺氏细胞,然后再用稀释50倍的正常小鼠血清处理.用这样处理的神经段植入对照组大鼠右侧腿内;其余神经段再用抗Col.Ⅳ抗血清处理后植入抗Col.Ⅳ组鼠内.术后5、7、10、15和30天取材,在移植神经段1、5和9mm处切片进行光镜和电镜观察,并作计数和统计学处理.实验结果显示:1.再生轴突数抗Col.Ⅳ组占对照组的百分比是:术后15天1和5mm处各为65%和4%;术后30天1、5和9mm处分别为94%、82%和57%.提示抗Col.Ⅳ组的再生轴突生长速度低于对照组.2.巨噬细胞数术后10天抗Col.Ⅳ组为对照组的1.7倍;术后15天为2倍;术后30天为3倍.提示Col.Ⅳ水平降低导致神经再生微环境的异常.结论;1.雪旺氏细胞基底膜成份Col.Ⅳ在早期神经再生中参与促进轴突的生长和成熟;2.Col.Ⅳ是神经微环境中不可缺少的物质,它对维持神经微环境的平衡起重要作用.文章对Col.Ⅳ的作用机制作了初步的分析和探讨.

The number of macrophages was 1.7 times that of the control group after 10 days of operation, twice as high as 15 days after operation, and three times as high as 30 days after operation. Conclusion 1. Schwann cells basement membrane composition Col.IV in early nerve regeneration involved in promoting axon growth and maturation; 2.Col.IV is an indispensable material in the neural microenvironment, it is to maintain the nervous microenvironment The balance plays an important role.The article makes a preliminary analysis and discussion on the mechanism of Col.Ⅳ.
