目的初步探讨FK506拮抗庆大霉素耳毒性的效果。方法通过分别检测庆大霉素损伤组与庆大霉素加不同剂量FK506(1mg、2mg)保护组实验动物听性脑干反应(ABR)阈值的变化,并结合耳蜗铺片琥珀酸脱氢酶(SDH)染色方法观察动物耳蜗形态学变化,观察FK506拮抗庆大霉素耳毒性的作用。结果实验动物的听觉生理检查显示FK506能够拮抗庆大霉素耳毒性,FK506保护组动物听阈明显低于庆大霉素损伤组(P<0.01),随剂量增加,保护作用越明显;同时形态结果显示FK506保护组外毛细胞缺失明显少于庆大霉素损伤组(P<0.01)。结论 FK506对庆大霉素引起的耳毒性有拮抗作用。
Objective To investigate the antagonistic effect of FK506 on gentamicin ototoxicity. Methods The changes of threshold of auditory brainstem response (ABR) in gentamicin and gentamicin plus different doses of FK506 (1mg, 2mg) protective group were detected respectively. Combined with cochlear melatonin succinate dehydrogenase (SDH) staining method to observe the morphological changes of the cochlea in animals and observe the effect of FK506 on gentamicin ototoxicity. Results The auditory and physiological examination of experimental animals showed that FK506 could antagonize gentamicin ototoxicity. The threshold of FK506-protected animals was significantly lower than that of gentamicin-treated rats (P <0.01), and the protective effect was more obvious with increasing dose. The loss of outer hair cells in FK506 group was significantly less than that in gentamicin group (P <0.01). Conclusion FK506 antagonizes gentamicin-induced ototoxicity.
