雷公藤红素对奥氮平所致大鼠糖脂代谢紊乱的改善作用

目的建立奥氮平诱导的糖脂代谢紊乱大鼠模型并研究雷公藤红素对糖脂代谢紊乱的作用。方法 24只SD雌性大鼠,按照体重随机分成对照组、模型组、2个剂量实验组。模型组给予奥氮平(1 mg·kg~(-1),ip,bid),实验组在建模的同时给予2个剂量(3,9 mg·kg~(-1)·d~(-1),ig,qd)雷公藤红素,对照组腹腔注射等量的生理盐水,记录4组大鼠的摄食量和体重的变化。28天后采血并将大鼠处死,测定血清总胆固醇(TC)、三酰甘油(TG)、高密度脂蛋白(HDL-C)、低密度脂蛋白(LDL-C)、空腹血糖(FBS)及胰岛素(INS)水平。结果 28 d后,对照组、模型组和低、高2个剂量实验组大鼠血清TG水平分别为(0.24±0.09),(0.50±0.11),(0.31±0.06),(0.31±0.03)mmol·L~(-1);HDL-C水平分别为(0.83±0.13),(0.33±0.24),(1.52±0.55),(1.78±0.24)mmol·L~(-1);INS水平分别为(32.40±16.70),(123.60±41.80),(38.31±8.53),(22.96±8.06)μU·mL~(-1),HOMA-IR分别为9.49±5.01,35.73±12.19,11.33±2.50,6.39±1.96。与对照组相比,模型组的TG水平显著升高,HDL-C水平显著降低,INS水平以及胰岛素抵抗指数(HOMA-IR)显著升高,差异均有统计学意义(均P<0.05);而LDL-C和FBS无明显变化。与模型组相比,2个剂量实验组血清TG、INS水平以及HOMA-IR显著降低,HDL-C水平显著升高,差异均有统计学意义(均P<0.01);而LDL-C、TC、FBS无显著差异。低、高2个剂量实验组间各指标均无显著性差异。结论雷公藤红素对奥氮平引起的糖脂代谢紊乱可起到明显的改善作用。 OBJECTIVE To establish a rat model of olanzapine-induced dyslipidemia and study the effect of tripterine on glucose and lipid metabolism disorder. Methods Twenty-four SD female rats were randomly divided into control group, model group and two dose groups according to body weight. The model group was given olanzapine (1 mg · kg -1, ip, bid). The experimental group was given 2 doses (3, 9 mg · kg -1 · d ~ (-1) 1), ig, qd) Tripterine, the control group, the same amount of saline injected intraperitoneally, recording the changes in food intake and body weight of 4 groups of rats. After 28 days, the rats were sacrificed and the rats sacrificed. The levels of total cholesterol (TC), triglyceride (TG), high density lipoprotein (HDL-C), low density lipoprotein (LDL-C), fasting blood glucose Insulin (INS) levels. Results After 28 days, the levels of TG in the control group, model group and low and high dose groups were (0.24 ± 0.09), (0.50 ± 0.11), (0.31 ± 0.06), (0.31 ± 0.03) mmol · L -1 and HDL-C levels were (0.83 ± 0.13), (0.33 ± 0.24), (1.52 ± 0.55) and (1.78 ± 0.24) mmol·L -1, respectively (32.40 ± 16.70), (123.60 ± 41.80), (38.31 ± 8.53) and (22.96 ± 8.06) μU · mL -1, HOMA-IR were 9.49 ± 5.01, 35.73 ± 12.19, 11.33 ± 2.50 and 6.39 ± 1.96. Compared with the control group, the levels of TG, HDL-C, INS and HOMA-IR in the model group were significantly increased (all P <0.05); There was no significant change in LDL-C and FBS. Compared with the model group, the levels of serum TG, INS and HOMA-IR in the two experimental groups were significantly decreased, and the levels of HDL-C were significantly increased (all P <0.01); while the levels of LDL-C, TC , FBS no significant difference. There was no significant difference between the two experimental groups in low dose and high dose. Conclusion Tripterine on olanzapine induced dyslipidemia can play a significant role in improving.
