目的研究老年期胃黏膜生长抑素含量和分布状况,探讨老化过程中生长抑素作为一种胃酸抑制因子存在的实际意义。方法选择不同年龄Wistar大鼠,雌雄随机。青年鼠(3月龄)20只,体重200~250g;老年鼠(24~30月龄)15只,体重400~450g。动物按规定月龄取血,分离血浆样本,提取胃黏膜组织,采用放射免疫分析法分别进行生长抑素含量测定。取胃窦新鲜组织,经冰冻切片进行生长抑素免疫组化染色。结果不同年龄组Wistar大鼠胃黏膜组织生长抑素具有不同的生理浓度,在老年鼠生长抑素含量呈高水平分布,与青年鼠比较有非常显著性差异(P<0.01)。生长抑素免疫组化染色显示,胃窦D细胞形状不规则,胞浆内布满生长抑素免疫复合物颗粒,老年鼠D细胞显色较深,其D细胞数量与免疫复合物颗粒高于青年鼠组。两组比较有显著性差异(P<0.05)。结论老年期胃黏膜D细胞数量增加,生长抑素分泌量增高,与其老年抑酸调节作用增强存在重要关系。
Objective To study the content and distribution of somatostatin in elderly patients with gastric mucosa and to explore the practical significance of somatostatin as a gastric acid suppression factor in the aging process. Methods Wistar rats of different ages were selected and randomly divided into male and female. Twenty young mice (3 months old) weighing 200-250 g and 15 aged rats (24-30 months old) weighing 400-450 g. Animals were taken blood at the required age, plasma samples were separated, gastric mucosa tissues were extracted, and somatostatin contents were determined by radioimmunoassay. Take fresh tissue of gastric antrum, somatostatin immunohistochemical staining by frozen section. Results Somatostatin in gastric mucosa of Wistar rats of different age groups had different physiological concentrations. There was a high level of somatostatin in aged rats, which was significantly different from that in young rats (P <0.01). The somatostatin immunohistochemical staining showed that the shape of D cells in gastric antrum was irregular and the somatostatin immunocomplex particles were covered in the cytoplasm. The D cells in old mice developed a deeper color and the number of D cells and immune complex particles were higher Young rats group. There was significant difference between the two groups (P <0.05). Conclusions The number of D cells in elderly patients with gastric mucosa increases and the secretion of somatostatin increases, which plays an important role in the enhancement of acid-suppressing effect in elderly patients.
